Abstract

The family Agromyzidae includes approximately 2,500 species worldwide, and their larvae feed by creating galleries in leaf and stem tissues. While most species are host plant specific, polyphagous species belonging to the genus Liriomyza are pests that cause serious economic losses in agriculture and horticulture and are on the quarantine lists of many countries. Larvae cause aesthetic and physiological damage by creating galleries in the leaf mesophyll tissue. In natural ecosystems, Liriomyza spp. is controlled by parasitoids belonging to the families Eulophidae, Braconidae, and Pteromalidae. Neochrysocharis formosa is an effective biological control agent on Liriomyza spp. in regions such as Japan, Türkiye and Italy. Female adults are 1.1–1.28 mm long and have a dark brown and metallic green-reflected head and mesosomes. Morphological similarities make species identification difficult using traditional methods. DNA barcoding provides rapid and reliable species identification using the mitochondrial COI gene region. In this study, the COI gene region of N. formosa samples collected from Aydın province was amplified by PCR and compared with the sequences in the NCBI GenBank database. Phylogenetic relationships among the samples were evaluated using Neighbor-Joining (NJ) analysis based on COI sequences. The phylogenetic analysis revealed that our samples were similar to the Italian reference sequence. DnaSP analyses of our samples show that they exhibit high haplotype diversity (Hd: 0.933) and low nucleotide diversity (π: 0.01645). The findings show that N. formosa was confirmed at the molecular level using the COI barcode region in accordance with the morphological description from the Turkish material, thus demonstrating that the species identification is supported bidirectionally (morphological + molecular).
