Abstract

Objectives: We investigated the changes occurring in the density of basic fibroblast growth factor (bFGF) immunoreactive (ir) cells 15 days after kainic acid (KA) injection in young and adult rat hippocampus. Study Design: A single injection of serum physiologic solution (control) or KA (7 mg/kg) was administered intraperitoneally to young (5 days) and adult (two months) Wistar albino rats. Fifteen days after the injection, and following intraaortic perfusion with the use of a fixative solution, the rats were decapitated, and the brains were removed. Mouse monoclonal bFGF antibodies were used to demonstrate bFGF immunoreactivity. By light microscopy, bFGFir cells were counted in all groups and evaluated statistically. Results: bFGF immunoreactivity was observed in the astrocytes in the hippocampus, and in the pyramidal neurons in the CA2 region both in the control and KA groups. Compared with the agematched controls, KAinjected rats demonstrated significantly increased numbers of bFGFir astrocytes in the hippocampus (young, p<0.0001 ; adult, p<0.001) and pyramidal neurons in the CA2 region (young, p<0.05; adult, p<0.001). Conclusion: Significantly increased bFGFir astrocytes and CA2 pyramidal neurons seen in KAadministered young and adult rats suggest the presence of a protective mechanism against epileptogenesis.
