Abstract
Biotic and abiotic stress factors cause significant problems in the cultivation of cucurbits. Among these factors, viral pathogens lead to substantial losses in both the quality and quantity of squash production. Viral agents can be easily transmitted from one plant to another through mechanical means, vector insects, seeds, and vegetative propagation. In addition to their rapid spread, the absence of chemical control against these agents complicates disease management and increases economic losses. One such agent is the Cucumber mosaic virus (CMV; Cucumovirus CMV). In this study, we investigated the presence of CMV in squash plants, which have considerable economic importance, cultivated in Çanakkale province and its districts. For this purpose, during the 2023 cultivation season, field surveys were conducted in squash production areas, and 25 samples were collected from plants showing viral or virus-like symptoms. The collected samples were tested for CMV infection using a serological method, double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), and a molecular method, Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR). The result of the analyses revealed that CMV infection was detected in four of the 25 samples (16%). The most typical symptoms observed in the confirmed CMV-infected samples were intense mosaic patterns and leaf deformations.
