Abstract

Walnut (Juglans regia L.) leaves are a plant material widely valorized in traditional medicine, cosmetic formulations, and the food supplement industry due to their rich phenolic composition. In this study, the phenolic composition profile of an extract (10000 ppm) obtained from walnut leaves using an ethanol–water solvent system was investigated by reversed-phase high-performance liquid chromatography with diode-array detection (HPLC-DAD). The analysis was performed at 300 nm signal and 500 nm reference wavelengths over a 0–65 min run time. A total of 16 peaks with retention times (tR) ranging from 3.266 to 35.712 min were identified in the resulting chromatogram: ascorbic acid, gallic acid, protocatechuic acid, catechin, hydroxybenzoic acid, gentisic acid, p-coumaric acid, rutin, ferulic acid, naringin, o-coumaric acid, neohesperidin, coumarin, resveratrol, quercetin, and trans-cinnamic acid. The peaks with the highest absorbance intensity were ascorbic acid (tR=3.266 min) and catechin (tR=6.440 min), both eluting in the early retention window (tR<10 min), which accounted for a considerable proportion of the total peak area. The findings indicate that the walnut leaf extract contains substantial levels of phenolic acids and flavonoids—particularly gallic acid, catechin, rutin, and quercetin—consistent with the general trends reported in the existing literature. These results support the potential use of walnut leaf extract as a natural phenolic source with antioxidant potential in cosmetic, food supplement, and functional product formulations.
