Abstract

The use of microwave in the electron microscopic fixation of nerve tissue is suggested to increase the quality and be economic in the use of time and chemicals. The purpose of this study is to achive a time gaining procedure, which includes the primary and secondary fixation for electron microscopy, by the use of microwave irradiation for the examination of normal and degenerated myelin in the central and peripheral nervous system. Rat brain and schiatic nerve were used for examination of normal myelin. Middle cerebral artery occlusion and full thickness schiatic nerve cutting were performed on rat to produce myelin degeneration. Tissues were fixed conventionally (3 hours) and with microwave irradiation for electron microscopic evaluation. Electron microscopical double fixation took 19 minutes in the microwave oven. Besides the quality of the slides were equal to the conventionally prepared ones. With these findings we concluded that microwave irradiation can be used for the light and electron microscopical examination of normal and degenerated myelin.
