Abstract

In this study, it is aimed to develop a method of multi residue screening analysis for detecting total 39 compounds (4 tetracycline, 4 macrolide, 8 sulfonamide, 1 diaminopyrimidine, 9 quinolones, 10 betalactams and 3 amphenicol derivatives) in poultry meat by likid chromatography tandem mass spectrometry (LC-MS/MS) system and to validate that method according to European Union Directive no 2002/657/EC. In order to detect the compounds, the extractions prepared by two extract process are given into LC-MS/MS system. In the first extraction, extracting the antibiotics from poultry meat was performed by Na2EDTA and 70 % methanol. Coarse particles were settled down by centrifugation, water was added while taking the upper phase, first injection was made into LC-MS/MS system after filtering. At the first extraction and first injection, 36 compounds of sulfonamide, tetracycline, quinolone, betalactam, macrolide and diaminopyrimidine derivatives were detected in the interval of 0.7 26.4 μg/kg. The second extraction was performed by water and ethyl acetate. Coarse particles were precipitated by centrifugation and evaporated until being dry while taking supernatant. The dried extraction was dissolved by water and n-hexan, centrifuged and second injection was made into LC-MS/MS system while filtering through lower phase. Three amphenicol derivative compounds were detected at the second extraction and second injection in the interval of 0.03 1.4 μg/kg.
