Abstract

In this study, the effects of heparin (1st group), caffein plus heparin (2nd group) and caffein plus calcium ionophore (3rd group) on in vitro capacitation of frozen-thawed bull semen were investigated. For this purpose, in the first group, three dosages of heparin (10, 25, 100 |-ig/ml) and three incubation time (5, 15, 60 min.) were examined. In the second group, 5, 10 and 15 inM concentrations of caffein combined with 0, 5, 10, 25 $mu$g/ml heparin and in the third group, 0, 5, 10 mM concentrations of caffein combined with 0.1, 0.2, 0.3 $mu$M calcium ionophore. After induction of capacitation by these agents, a dose of 100 mg/ml lysophosphatidylcholine (LPC) was added to medium to induce acrosome reaction for a period of 15 minutes. To determine the reaction and viability, spermatozoa were stained by dual staining method using trypan blue and Giemsa. Four categories of spermatozoa were identified and counted: 1- Intact acrosome, live; 2- Intact acrosome, dead; 3- Detached acrosome, live; 4- Detached acrosome, dead. In the first, second and third groups the highest results were 25.466% (100 fig/ml heparin, 60 min.), 28.440% (15 mM caffein plus 25 $mu$/ml heparin) and 29.908% (10 mM caffein plus 0,3 $mu$M calcium ionophore), respectively. The mean proportions of capacitated spermatozoa increased as the heparin dosage and incubation period increased in the first group. A synergistic effect of caffein plus heparin and caffein plus calcium ionophore were observed in our study. The total dosages of capacitating agents increased, mean proportions of capacitated spermatozoa increased in the second arid third groups.

Özet

Çalışmada, heparin (1. grup), heparin+kafein (2. grup) ve kafein+kalsiyum ionoforun (3. grup) in vitro spermatozoon kapasitasyonu üzerine etkileri incelendi.
