Abstract

Purpose: Acetone is widely used as an industrial solvent and chemical intermediate. Most products containing acetone are used by industry, also some acetone products, such as nail polish remover, paint remover, glue, and cleaning agents, are used by consumers. It has been detected in groundwater and drinking water. Lipid peroxidation is associated with a wide variety of toxicological effect, including decreased membrane fluidity and function, impaired mitocondrial and golgi apparatus functions, and inhibition of enzymes. In this in vitro study, we aimed to investigate if acetone, a widely used agent and so having a high risk of contact, causes lipid peroxidation at erythrocyte membrane or not, and if it has any hemolytic effect. Methods: Blood samples that were obtained from two weeks old stored blood were put into tubes with anticoagulant and at different concentrations acetone was added these tubes. After 48 hours, plasma and erythrocyte samples were seperated and studied aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and iron levels in the plasma. It was evaluated the oxidative state of erythrocytes by studied MDA levels which is the indicator of lipid peroxidation. Result: AST, LDH and iron levels in acetone added tubes were significantlly higher than the controls, their elevated levels was correlated with increased concentrations of acetone. No significant difference was observed between the study and control samples about MDA levels, a lipid peroxidation marker. Conclusion: It was concluded that acetone may cause hemolysis of erythrocytes with its toxic effect in vitro conditions and this is not related with lipid peroxidation in acute period (48 hours) but may relate with lipophilic and solvent effect of acetone on erythrocyte membrane.
