Abstract

Nowadays, under appropriate conditions, in vitro biopharmaceutical investigations can be used instead of in vivo bioequivalence studies by legislating the regulations considering the human health and the ethical rules. Therefore, it is important to examine the solubility, permeability, and drug release from the pharmaceutical dosage form to reduce the high cost in vivo bioequivalence studies in healthy volunteers. The Biopharmaceutics Classification System (BCS) is basically a classification system based on the solubility and permability criteria. Depending on these criteria, active pharmaceutical substances are categorized into four main classes: BCS Class 1-high soluble/high permeable drugs, BCS Class 2-low soluble/high permeable drugs, BCS Class 3-high soluble/low permeable drugs, BCS Class 4-low soluble/low permeable drugs. BCS, when evaluated with dissolution is an important step in the waiving from in vivo bioequivalence studies for generic drugs. In previous years, various health authorities have specified different criteria for waiving conditions, but in current regulations, authorities have adopted a common approach to minimize the differences. Thus, BCS has been accepted by the health authorities of many countries such as Food and Drug Administration and European Medicine Agency and has been included in current guidelines. In our country, the Ministry of Health Turkish Medicines and Medical Devices Agency takes into account the biowaiver applications. In this study, the approaches of various health authorities to the concept of biowaiver and the BCS applications in national and international guidelines have been extensively discussed.

Özet

Günümüzde, etik kurallar ve insan sağlığı göz önünde bulundurularak yapılan güncel yasal düzenlemelerle, uygun koşullar sağlandığında in vitro biyofarmasötik değerlendirmeler, in vivo biyoeşdeğerlik çalışmalarının yerine kullanılabilmektedir.
