Abstract

Purpose: Epilepsy is a complex neurological disorder resulting from abnormal neuronal activity and involving multiple neurotransmitter mechanisms, including the dopaminergic system. This study aimed to evaluate the effects of the dopamine D2 receptor agonist pramipexole on electrophysiological activity in a penicillin-induced rat model of epilepsy and to investigate its possible interaction with valproate. Methods: Thirty-five male Wistar albino rats were used and randomly divided into five groups: control, pramipexole 1 mg/kg, pramipexole 3 mg/kg, valproate 300 mg/kg, and valproate + pramipexole 3 mg/kg. The epilepsy model was induced by intracortical penicillin injection. Electrocorticogram (ECoG) recordings were obtained for 180 minutes, and spike-wave frequencies and amplitudes were analyzed. Data were evaluated using Kruskal–Wallis and Mann–Whitney U tests. Results: A significant reduction in spike-wave frequency was observed in the group treated with 3 mg/kg pramipexole (p=0.012). Both valproate (p<0.001) and the valproate + pramipexole (p<0.001) groups exhibited marked anticonvulsant effects; however, no additive interaction between the two drugs was detected. Pramipexole at 1 mg/kg did not show a significant anticonvulsant effect. Conclusion: Pramipexole at a dose of 3 mg/kg exerts anticonvulsant effects in a penicillin-induced epilepsy model. No additional therapeutic benefit was observed when combined with valproate. These findings suggest that dopamine D2 receptors play an important role in the regulation of epileptic activity and that the dopaminergic system may contribute to epileptogenesis.
