Abstract

Andrographis paniculata is a traditionally utilized medicinal herb for its potent anti-inflammatory activity, mainly attribute andrographolide, the major diterpenoid compound. However, inadequate water solubility and lower bioavailability of its active components limit its pharmaceutical applications. This research intended to design and enhance a stable Andrographis paniculata extract nanoemulsion using a low-energy emulsification method and the Simplex Lattice Design approach. Oleic acid, polysorbate 80, and propylene glycol were used as the oil phase, surfactant, and cosurfactant, respectively. The optimized formulation (oleic acid:Smix ratio of 4:19) produce a droplet size of 132.678 nm, a polydispersity index of 0.233, and zeta potential of -23.2 mV. The nanoemulsion maintained thermodynamic stability under centrifugation, heating-cooling, and freeze-thaw cycles with no notable changes in its characteristic. TEM anlysis confirmed spherical, uniformly distributed droplet without agglomeration, while FT-IR spectroscopy verified andrographolide incorporation withouts significant chemical interaction. The result indicate that the low-energy method successfully produced a stable, homogeneous, and physically compatible Andrographis paniculata extract nanoemulsion with promising potential for pharmaceutical applications, particularly topical delivery.
