Abstract

This study evaluated the in vitro antibacterial potential of hydrosol obtained from Arum dioscoridis (Tirşik) against bacterial pathogens isolated from lizardfish ( Saurida lessepsianus ) collected in İskenderun Bay. Bacterial isolates were identified using the Phoenix 100 ID/AST automated identification system (Becton Dickinson, Sparks, MD, USA). Antibacterial activity was assessed by the agar well diffusion assay, and inhibition zone diameters were recorded at four hydrosol concentrations (100%, 75%, 50%, and 25%, v/v). The tested isolates comprised Staphylococcus aureus , Klebsiella pneumoniae, Enterobacter cloacae, Pseudomonas aeruginosa, Escherichia coli, and Proteus mirabilis . The hydrosol exhibited the most pronounced activity against S. aureus , producing inhibition zones up to 25 mm at 100% concentration and maintaining detectable inhibition even at 25% concentration. In contrast, Gram-negative isolates displayed lower susceptibility, with inhibitory effects predominantly observed at higher concentrations. Notably, no antibacterial activity was detected against E. coli and P. mirabilis at any tested concentration. Overall, the findings highlight A. dioscoridis hydrosol as a promising natural antibacterial agent, particularly against Gram-positive fish-associated pathogens, and support further investigations including in vivo validation for potential aquaculture applications.

Özet

Bu çalışmada, İskenderun Körfezi’nden temin edilen İskarmoz ( Saurida lessepsianus ) balıklarından izole edilen bakteriyel patojenlere karşı Tirşik ( Arum dioscoridis ) bitkisinden elde edilen hidrosolün in vitro antibakteriyel etkinliği araştırılmıştır. Bakteri izolatlarının tanımlanması Phoenix 100 ID/AST (Becton Dickinson, Sparks, MD, ABD) otomatik tanımlama sistemi ile gerçekleştirilmiştir. Hidrosolün antibakteriyel etkisi agar well difüzyon yöntemi kullanılarak belirlenmiş, dört farklı konsantrasyonda (%100, %75, %50 ve %25) inhibisyon zon çapları ölçülmüştür.
