Abstract

Objective: Fat mass and obesity-associated (FTO) protein is an epitranscriptomic regulator that plays crucial functions in the regulation of energy metabolism, cellular stress response, and gene expression through demethylation N6-methyladenosine (m⁶A) and N6,2′-O-dimethyladenosine (m⁶Am) modifications on RNA. The endoplasmic reticulum-associated degradation (ERAD) pathway plays a critical role in maintaining cellular proteostasis by eliminating misfolded or defective proteins. However, the potential relationship FTO and ERAD pathway remains insufficiently characterized. Methods: In this study, the effects of wild-type FTO (WT-FTO) protein overexpressing on the ERAD-associated protein levels were investigated in neuronal SH-SY5Y cells. A tetracycline-inducible stable expression system was employed. Protein levels were assessed by Western blotting, and band intensities were quantified using ImageJ and GraphPad Prism. Statistical analyses were performed using unpaired two-tailed Student’s t-test. Results: WT-FTO expression was successfully induced, resulting in an approximately four-fold increase in FTO protein levels. Overexpression of WT-FTO did not significantly alter GRP78/BiP levels, a canonical ER stress marker. However, significant increases were observed in VCP/p97 (~6-fold) and GRP58/ERp57 (~2-fold) protein levels (p<0.05). Conclusion: These findings suggest that WT-FTO overexpression may be associated with selective modulation of ERAD-related components without clear evidence of a classical ER stress response. The data indicate a potential link between epitranscriptomic regulation and proteostasis; however, further functional studies are required to clarify the underlying mechanisms.
