Abstract

Comparative Analysis of Populations of Three Species within the Psephellus aucherianus species somplex in terms of matK to determine the level of genetic variation and the boundaries of the taxa of Psephellus sintenisii, Psephellus yusufeliensis, and P. aucherianus, which are part of the Psephellus aucherianus complex within the genus Psephellus, a partial region of the matK regions (chloroplast DNA) were amplified and sequenced. According to the data obtained, it was determined that the population Eriç, belonging to the P. aucherianus, represents an ancestral lineage from an evolutionary perspective, and that population of each species has its own unique haplotype diversity over time. A total of 14 haplotypes were identified in the six populations within this complex. Three of these haplotypes were specific to the P. sintenisii, while three were specific to the P. yusufeliensis. The remaining eight haplotypes belonging to the P. aucherianus and its populations, and one of them is shared between the Bağıştaş and Eriç (Erzincan) populations. Thus, it is revealed that these two populations are genetically much more closely related compared to the others. Furthermore, the levels of genetic variation and differentiation, calculated according to the identified haplotypes, were found to be quite variable among themselves, providing us with information for developing appropriate conservation strategies. Consequently, priority should be given to the conservation of P. sintenisii, a species with relatively low genetic diversity. In addition, considering the levels of genetic variation and allelic diversity of the other two species, the moderate values determined indicate that they are also at risk, although not to the same extent as the first species.
