Abstract

Objective: Alcohol is a toxic substance that leads to tissue and organ damage through an inflammatory cascade involving numerous cytokines, chemokines, and reactive oxygen species. Given that chronic inflammation is recognized as a central factor in various diseases, recent studies have focused on foods rich in polyphenols with anti-inflammatory and immunomodulatory properties. This study aims to investigate the protective and therapeutic effects of resveratrol, a widely studied polyphenol, on alcohol-induced human umbilical vein endothelial cell (HUVEC) lines. Material and Method: In our study, 4 different experimental groups were formed as control group, alcohol exposure group, resveratrol group, alcohol and resveratrol co-administered group. The doses to be applied were determined as the inhibitory concentration 30 (IC30) dose (4.4%) for alcohol and the highest non-toxic dose (20 µM) for resveratrol according to the results obtained from cytotoxicity experiments. The study was conducted on human umbilical vein endothelial cell (HUVEC) lines. The parameters examined, including Interleukin 18 (IL-18), C-reactive protein (CRP), and caspase-3 activity, were measured using ELISA kits. Result and Discussion: Resveratrol supplementation has been shown to provide protection against inflammation by significantly reducing the levels of the inflammatory cytokine interleukin 18 (IL-18) in response to alcohol (%20.31, p<0.05). Additionally, resveratrol significantly reduced C-reactive protein (CRP) levels (%17.41, p<0.05). It was also demonstrated that alcohol significantly increased caspase-3 activity in HUVEC cell lines, while resveratrol administration effectively inhibited this increase (%12.75 p<0.05). It is suggested that resveratrol reduces CRP levels and mediates caspase-3 inhibition by alleviating alcohol-induced oxidative stress. Given that resveratrol has not shown any adverse effects on human health and possesses protective properties against oxidative stress, it is considered that it could be used as a nutraceutical supplement in conjunction with pharmaceutical treatments.

Sonuç ve Tartışma: Resveratrol takviyesinin alkole karşı, inflamatuar sitokinlerden interlökin 18 (IL-18)’i önemli ölçüde azaltarak inflamasyona karşı koruma sağladığı gösterilmiştir (%20.31, p<0.05).
