Enteral beslenme ürünlerindeki vitamin B2 miktarının HPLC ile belirlenmesi

Özet
Amaç: Enteral beslenme ürünlerindeki vitamin B2 (riboflavin) miktarının tayinine yönelik floresans dedeksiyonu (FL/250-546 nm) ile kolay, spesifik ve duyarlı, yüksek basınçlı sıvı kromatografi (HPLC) yönteminin uygulanabilirliğini tespit etmektir. Yöntem: Toplam 48 adet enteral beslenme ürününün vitamin B2 miktarlarının belirlenmesi için HPLC yöntemi kullanılmıştır. Kromatografik sistem; C-18 kolon (250x4,60-5micron) ve 250-546 nm'ye ayarlı floresans dedektör kullanılarak gerçekleştirilmiştir. Mobil faz olarak 1 ml/dk akış hızında hekzansülfonik asit sodyum tuzu içeren tamfton ve metanol (% 75:25) kullanılmıştır. İstatiksel değerlendirme, iki eş arasındaki farkın önemlilik testine göre yapılmıştır. Bulgular: B2 vitaminin alıkonma süresi yaklaşık 14 dakika olarak saptanmıştır. Enteral beslenme ürünlerindeki vitamin B2 miktarları ise 0,11 -0,40 mg/100 mi arasında tespit edilmiştir. Bu tespit edilen değerlerin, ürün etiketlerindeki değerlere uygunluk gösterdiği belirlenmiştir. Sonuç: Enteral beslenme ürünlerindeki vitamin B2 miktarlarının belirlenmesinde HPLC-FL (HPLC-Floresans Dedektör) kullanılabileceği görülmüştür. Ayrıca bu metodun duyarlı, uygulama süresi ve kolaylığı açısından avantajlı bir yöntem olduğu da düşünülmüştür.
Abstract
Objective: The aim of this study is to determine the applicability of fluorescence detection (FL/250-546 nm) method by the simply, specific and sensitive high pressure liquid chromatography (HPLC) for quantitative estimation of vitamin B2 (riboflavin) levels in enteral nutritional products.Method: HPLC method was used for quantitative determination of vitamin B2 in total 48 enteral nutritional products. The chromatographic system was done on a C18 column (250x4,60-5micron) with fluorescence detection at 250-546 nm. Buffer containing hexanesulphonic acid sodium salt and methanol (75:25) was used as the mobile phase at a flow rate of 1 ml/min. The results were evaluated using the paired samples test. Results: The retention time of vitamin B2 was determined as ca. 14 min. The concentration of vitamin B2 of enteral nutritional products ranged from 0.11 to 0.40 mg/100 ml. The determined values showed similarity with the labelling values of the products. Conclusion: It was showed that HPLC-FL (HPLC-Fluorescence Detector) can be applied for the quantitative determination of vitamin B2 levels in enteral nutritional products. This had the advantages of improved simplicity, sensitivity and shorter application times.
