Abstract

Colorectal cancer is responsible for approximately 10% of cancer-related deaths today. Dysregulation of signaling cascades causes colon epithelium to develop into malignant tumors. One of these cascades, the TGF-β (Transforming growth factor beta) signaling pathway, can be dramatically changed to promote metastasis and the epithelial-to-mesenchymal transition (EMT) in later stages of the illness, even though it functions as a tumor suppressor in early neoplastic processes. ADAMTS-5, which has a potential role in different types of cancer, is an extracellular metalloproteinase that degrades extracellular matrix components such as aggrecan and versican and plays a role in embryonic development. It has been demonstrated that overexpression of ADAMTS-5 stimulates the growth of human glioblastomas, while downregulation or hypermethylation of ADAMTS-5 inhibits a variety of cancers, such as gastric cancer, human colorectal cancer, and melanoma. In our study, the effects of hypoxia and TGF-β conditions on the protein and mRNA levels of the ADAMTS-5 gene were determined in HT-29 cells, a colon cancer cell model. CoCl2-induced chemical hypoxia model was used to create hypoxic conditions. In our work, the cytotoxic effects of TGF-β and CoCl2 applied to the cells were determined by the MTT method. Real-time PCR was used to assess the effects of TGF-β cytokine and CoCl2 administered to the cells separately and combined on the ADAMTS-5 mRNA level, and Western blot was used to assess the effects on the protein level. Considering our study's findings, ADAMTS-5 was shown to increase with hypoxic regulation. It was determined that TGF-β cytokine significantly reduced ADAMTS-5 expression.
