Abstract

Objective: This study aimed to investigate possible ototoxicityassociated with topical rifamycin application viaelectrophysiological tests and ultrastructural examinations. Methods: Electrophysiological assessment was performedwith tympanometry, auditory brainstemresponse (ABR), and distortion product otoacousticemission (DPOAE) measurements. This study wasconducted on 40 ears of 20 guinea pigs that weredetected to have normal hearing thresholds. The animalswere randomly assigned to three groups: Group1 (n=12) received 0.1 mL rifamycin, Group 2 (n=8)received 0.1 ml gentamycin, and Group 3 (n=20) received0.1 mL physiological saline. The antibiotics andsaline solutions were administered via intratympanicinjections. After five injections every other day, electrophysiologicaltests were performed again on the15th day. After electrophysiological measurements, the temporal bones of all guinea pigs were preparedfor ultrastructural examinations and the cochlear surfacemorphology was examined by scanning electronmicroscopy (SEM).Results: The animals in group 3 did not show a statisticallysignificant change in their DPOAE signal/noise ratio(SNR) or ABR thresholds (p>0.05). In groups 1 and2, the reduction in the DPOAE SNR and the increase inthe ABR threshold were statistically significant (p

Özet

Amaç: Bu çalışmada topikal rifamisin uygulamasının olası ototoksisitesi elektrofizyolojik testler ve ultrastrüktürel incelemeler ile araştırıldı.Yöntemler: Elektrofizyolojik değerlendirme timpanometri, işitsel beyinsapı yanıtları (ABR) ve distorsiyon ürünü otoakustik emisyon (DPOAE) ölçümleri ile gerçekleştirildi. Normal işitme eşiği tespit edilen 20 adet kobayın 40 kulağı çalışmaya dahil edildi ve rastgele olarak 3 gruba ayrıldı. Birinci gruba (n=12) 0.1mL rifamisin, 2. Gruba (n=8) 0.1 mL gentamisin, 3.
