Abstract

It is claimed that Polymerase Chain Reaction (PCR) could be used for the rapid diagnosis of tuberculosis in fresh tissue, paraffin blocks, sputum, gastric aspirate and cerebrospinal-fluid. In order to test this claim, sections obtained from paraffin blocks of 40 cases , histopathologically diagnosed as tuberculosis, were stained with Ziehl-Neelsen Method and also PCR was applied to all specimens. In six cases, Mycobacterium tuberculosis was detected with Ziehl-Neelsen stain, but none of the specimens was positive with PCR method. As control, the sputa of 96 patients tentatively diagnosed as tuberculosis with clinical findings and tuberculin skin test were studied with PCR and a positivity in 80 (83%) was obtained. We obtained positive results in 80 (83%) of 96 patients. It is concluded that PCR is not a sensitive method in paraffin blocks of tissues although it is routinely used in sputum and fresh tissues.
