Abstract
The cytotoxicity of 6-thioguanine in circulating T lymphocytes from healthy donors has been determined. In this paper, we demonstrated that 6-thioguanine inhibits the lymphocyte proliferation and leads to slight or severe structural chromosome abnormalities. These are premature chromosome condensation (PCC) long arm (q), short arm (p) and more progressive (eyeglass chromosome)telomeric chromosome arm associations, tremulous, dicentric and accessory chromosome deformity - abnormalities were established. Özet
Araştırmada, bir pürin analoğu olan ve primer olarak çocukluk çağı akut lösemide kullanılan 6-thiogua-nin'in insan T-lenfositler üzerine in vitro etkisi araştırılmıştır. Ajanın, 4-20 uM/öml dozlarında lenfosit proliferasyonu üzerinde sitotoksik bir etkisinin olduğu görülmüştür. Mitotik indeks üzerine olan bu olumsuz et¬kisinin yanısıra metafaz kromozomlarında da; gap, akse¬suar kromozom, titrek yada prematür kromozom, 'disent-rik kromozom ve bazı grup kromozomlarda bazen göz¬lük kromozom yapısına varan telomerik assosiye kromo¬zomlara neden olmaktadır. Yine ajana bağlı Qiemsa bo¬yamada, metafaz kromozomların homojen olmayan böl¬gesel boyanma farklılıkları gösterdikleri saptanmıştır.
