Abstract

In this study, the effects of UV-C stress –being one of the abiotic stress factors– on in vitro shoot development and antioxidant enzymes of Amsonia orientalis cultured in the presence of 6benzylaminopuine were examined based on the analysis of morphometric parameters, antioxidant enzymes, MDA (malondialdehyde) and H2O2 contents of in vitro shoots. All tested UV-C radiation dosages; 3.47 (15 min), 6.94 (30 min) and 13.87 (60 min) kJ m-2 reduced shoot length and shoot number compared to the control (non UV-C-applied). In the presence of 6-benzyaminopurine, radiation-sourced stress decreased shoot development but did not totally inhibit. UV-C radiation exerted different effects on antioxidant enzymes. It was found that SOD (superoxide dismutase) activity was reduced with respect to increasing radiation doses while POD peroxidase activity was enhanced. CAT activity was not significantly affected by radiation. Long-term UV-C exposure has been shown to increase similarly both H2O2 and MDA levels. Electrophoretic analysis showed that there are three SOD isozymes (Cu/Zn-SOD, Fe-SOD ve Mn-SOD) found and Cu/Zn-SOD was totally inhibited under all UV-C treatments. The results indicate that peroxidases play more important role than the other antioxidant enzymes in the plant’s defense mechanism against UV stress.
