Abstract

OBJECTIVE: Sexual assault is an important case of forensic science. The mostly used sample for investigation of these cases issemen. This investigation includes the examination of biochemicalmarkers present in semen such acid phosphatase and prostatespecificantigen, and also the assessment of the existence ofsperm in the sample using microscopy. In our study, we aimed toestablish the sensitivity and the selectivity of the acid phosphatasetest, PSA test and microscopy assessment for the investigation ofthese cases.METHODS: In our method, semen was examined either after normalincubation conditions or change in the condition of humidity. The semen samples used were mixed either with biologicalor non-biological samples at range of dilutions and used acidphosphatase test, PSA test and microscopy assessment.RESULTS: Acid phosphatase give positive results in semen, vaginalfluid and male urine, and PSA test give positive results notonly semen but also male urine. |t is also found that change inconditions of incubation [humidity] caused reduced detection ofsemen in the samples.CONCLUSION: Acid phosphatase test and PSA test alone are notspecific for the examination of semen so that microscopy assessmentis required for all the stains semen suspects. This study hasexamined the semen under the different conditions of incubationby acid phosphatase test, PSA test and microscopy assessmentused for investigation of sexual assault cases. Since there hasbeen no such report including all these available tests and differentconditions for samples, our study to our knowledge is thefirst in the field.
