Abstract

The protective effect of melatonin and the combination of vitamin C and vitamin E on the brain tissue of rats treated with chlorpyriphos-ethyl (CE) was investigated. The experimental groups were as follows: Control group (C), CE treated group (CE), vitamin E plus vitamin C treated group (Vit), melatonin treated group (Mel), vitamin E plus vitamin C plus CE treated group (Vit+CE), and melatonin plus CE treated group (Mel+CE). Vitamins and melatonin were administered to appropriate groups six days. CE was applied only fifth and sixth days. In CE group, comparing with C group, thiobarbituric acid reactive substances was found to increase, whereas antioxidant potential was decreased (p<0,10) in brain tissue homogenates. Superoxide dismutase and glutathione peroxidase activities were high in CE group, comparing with C group (p<0,10). These results suggest that CE may cause oxidative stress in rat brain tissues, this oxidative stress may involve in CE toxicity, and melatonin and the combination of vitamin C and vitamin E may decrease these toxic effects caused by CE.

Özet

Chlorpyriphos-ethyl'e (CE) maruz kalan ratlarin beyin dokularinda melatonin ile vitamin C ve vitamin E kombinasyonunun koruyucu etkileri arastirildi. Deney gruplari su sekilde organize edildi: Kontrol grubu (C), CE verilen grup (CE), vitamin E + vitamin C verilen grup (Vit), melatonin verilen grup (Mel), vitamin E + vitamin C + CE verilen grup (Vit+CE) ve melatonin + CE verilen grup (Mel+CE). Ilgili gruplara alti gün vitaminler ve melatonin verilirken besinci ve altinci gün CE verildi. Beyin dokusu homojenatlarinda CE grubunda C grubuna göre tiobarbitürik asit reaktif substans yüksek iken antioksidan potansiyel düsüktü (p<0,10). Süperoksit dismutaz ve glutatyon peroksidaz aktivitelerinde CE grubunda C grubuna göre düsük bulundu (p<0,10). Sonuçta, CE'nin rat beyin dokusunda oksidatif strese neden oldugu, bu oksidatif stresin CE toksisitesinde rol oynayabilecegi ve melatoninin ile vitamin C ve vitamin E kombinasyonunun CE'nin beyin üzerine toksik etkilerini anlamli olarak azaltabilecegi kanaatine varildi.
