Abstract

In recent years sputum induction has been used as a non-invasive research tool providing important information about airway inflammation. The aim of this study was to evaluate both the tolerability, safety and success of sputum induced and also the changes in sputum composition between the exacerbation and stable period of the chronic obstructive pulmonary disease (COPD). Sputum induction was performed in 15 patients with moderate to severe COPD during stable period and exacerbation. As a control group, 20 healthy adults (10 smokers and 10 nonsmokers) were included the study. Samples were analyzed for total cell count, viability and differential cell count. Finally, sputum analysis has given evidence for increased numbers of neutrophils in COPD patients compared to normal subjects. Whereas mainly macrophages were seen in healthy volunteers. In COPD patients, the mean total cell count was 16.7 ± 9.3 x 106 cells/mL, proportions of neutrophils was 91 ± 7.1%, proportions of macrophages was 8.8 ± 7.1% and cell viability was 87.8 ± 6.5% during exacerbation; the mean total cell count was 6.6 ± 7.1 x 106 cells/mL, proportions of neutrophils was 67.9 ± 20.2%, proportions of macrophages was 31.0 ± 20.4% and cell Bilgiviability was 81.6 ± 9.2% during stable period. The mean total cell count of healthy nonsmoker subjects was 2.4 ± 1.4 x 106 cells/mL, proportions of macrophages was 69.5 ± 18.3%, proportions of neutrophils was 29.7 ± 17.3% and viability 77.9 ± 9.6%; in healthy smokers the mean total cell count was 3.1 ± 1.5 x 106 cells/mL, proportions of macrophages 59.6 ± 23.2%, proportions of neutrophils was 40.1 ± 23.2% and viability 82.5 ± 9.3%.
