Abstract

Objectives: To investigate the effects of antioxidant vitamin and mineral treatment on outer retinal layers, Bruch’s membrane and choroid in phototoxic retinopathy model formed for age-related macular degeneration in rabbits. Material and Method: Twenty-four eyes of 12 New Zealand rabbits were used in the study. The rabbits were divided into four groups and left eyes were used as a control group. Right eyes of the 1st group were exposed to 2.5 Watt/cm2 light of operating microscope for 1 hour. In the 2nd group, 25 mg vitamin E, 200 mg vitamin C and 2.2 mg selenium were administration intramuscular daily for one week and right eyes were exposed to light in the same way. Right eyes of the 3rd group were exposed to light in the same way by attaching a blue filter to the microscope. Right eyes of the 4th group were exposed to light by applying a blue filter to the microscope after intramuscular application. One week after the procedure, all eyes were enucleated and examined with light and electron microscopes. Results: In light microscopy, losses and breaks in photoreceptor layer, retina pigment epithelium irregularity, drusen-like formations and lipoid deposition under retina pigment epithelium and fibroblast cells in the choroid layer were observed in the 1st and 3rd groups. In the 2nd and 4th groups the outer nuclear layer and retina pigment epithelium were more regular. Also, drusen or lipoid-like structures were not observed. Electron microscopy showed similar thickening and breaks of Bruch’s membrane, retina pigment epithelium irregularity and increased vacuoles, and intense collagen synthesis under retina pigment epithelium in the 1st and 3rd groups. However, these effects were very limited in the 2nd and 4th groups. Conclusion: Intense exposure to white and blue light causes retinal toxicity. This situation can be reduced by prior antioxidant vitamin and mineral intake.
