Abstract

Purpose: Self-prepared fortified antibiotic eye drops are frequently used to treat severe ocular infections. The potency and stability of these eye drops may change due to storage conditions including temperature, concentration and duration. In this study, in vitro potency and stability of vancomycin, cefazoline, amikacin and ceftriaxone at +4 degrees C in refrigerator and at +24 degrees C, were investigated. Material and Methods: Stock solution of cefazolin, vancomycin, amikacin and ceftriaxone were prepared from parenteral antibiotics by reconstituting lyophilized powder with artificial tear vehicle to concentration of 100 mg/ml, 50 mg/ml, 20 mg/ml and 133 mg/ml, respectively. For antimicrobial potency, cefazoline and vancomycin against Staphylococcus aureus, amikacin and ceftriaxone against Pseudomonas aureginosa and cefazoline, amikacin and ceftriaxone against Escherichia coli were tested. On days 0, 1 to 7, 10, 15 and 20, the minimum inhibitory concentration (MIC) were measured. We also examined PH for stability of these drops. Results: Cefazoline and vancomycin exhibited no loss of antibacterial potency during the 20 days at +4 degrees C and at 24 degrees C. However, amikacin was lost antibacterial potency after 15 days. It was found that storage temperatures had no effect on the potency. In all of the antibiotics, PH did not change and contamination was not detected during the study period. Conclusion: Fortified ophthalmic antibiotic solutions can be used for longer than 7 days at 24 degree C without any loss of its activity.
