Abstract

This study was carried out to observe the effect of ovariectomy on arginase (L-arginine amidinohydrolase, EC 3.5.3.1) activities of liver and kidney in normal and diabetic rats and to find out if there is a protective effect of estrogen and vitamin E administration on arginase activity in ovariectomized rats. Seventy Wistar albino rats were divided into seven groups as follows: 1) Control group, 2) Ovariectomy group, 3) Ovariectomized and Estradiol 17-p-treated group, 4) Ovariectomized, Estradiol 17-p- and vitamin E-treated group, 5) Ovariectomized, diabetic group (Streptozotocin to ovariectomized rats was intraperitoneally (IP) administered at a dose of 45 mg/kg body weight), 6) Ovariectomized, diabetic and Estradiol 17-P-treated group 7) Ovariectomized, diabetic, Estradiol 17-p- and vitamin E- treated group. Estradiol 17-p (E2) was administered subcutan at a dose of 40 /<g/kg, vitamin E (dl-a-tocopheryl acetate) was done intra periton at a dose of 100 mg/kg over 24 h for 28 days. Liver arginase activity was not affected by ovariectomy, but kidney arginase activity significantly decreased (P<0.001). Estradiol 17-p- and vitamin E- administration of ovariectomized rats did not change the activities. Liver arginase activity of ovariectomized diabetic rats was found to be significantly increased (P<0.001). In the ovariektomized diabetic group the arginase activity increased significantly (P<0.001) and by the administration of Estradiol 17-P alone in ovariektomized diabetic rats, arginase activity in the liver did not change, whereas administration of Estradiol 17-P and vitamin E combination made the activity level to turn to normal levels. In the ovariectomized diabetic group kidney arginase activity did not change whereas adminisration of Estradiol 17-P increased the arginase activity significantly (P<0.001). We suggest that proline, glutamate, and polyamine synthesis in kidneys may decrease by lowering kidney arginase activity in ovariectomy and that may cause to begin the kidney problems.
