Abstract

Serological methods provide quick results and give considerable contribution to the etiological diagnosis of infections. ELISA and fluorescent-antibody techniques are the serological methods most frequently used. The kits used in these assays are not produced in Turkey. The high expense of these imported kits restricts their routine utilization for experimental and diagnostic purposes. We prepared various bacterial, viral and parasitic antigens and used them in ELISA and indirect fluorescent-antibody (IFA) tests. We prepared adsorbed mycobacterial sonicated antigens (SAMTA) and the mycobacterial cell-wall antigen in the serological diagnosis of tuberculosis by ELISA. Sensitivity and specifity were 88.8% and 96.5%, respectively for SAMTA and 83.3% and 90%, respectively for mycobacterial cell-wall antigens For diagnosis of hydatid cyst disease, antigens were prepared from the cyst fluids of infected sheep and human for both assays. The sensitivities were 85.7%-89.2% and 75% by ELISA and IFA, respectively, and specifities were 95%-100% and 96.2% respectively In order to diagnose rubella, we utilized antigens derived from the vaccine strain. The sensitivity and specifity of ELISA were 100% and 63.6%, respectively. The figures for IFA were 96% and 63.6%, respectively. Our results show that the antigens prepared in our laboratory provide comparable sensitivity and specifity with commercial kits. Consequently we believe that our antigens can be used in the diagnosis, screening and epidemiological surveys of infectious diseases.
