Abstract

Aim: Squamous cell carcinoma is most frequent among oral cancers, which causes tumor progression in tongue. Most of the oral cancers could be diagnosed at a relatively late term. New diagnostic and therapeutic approaches are required for better treatment of patients. This study aimed to develop xenograft oral squamous cell carcinoma mice model, which could potentially support better diagnosis and treatment of patients. Materials and Method: Squamous cell carcinoma cell line OSC-19 was purchased from Japanese Collection of Research Bioresources (JCRB). Culture conditions were optimized accordingly, and growth and proliferation of cells were investigated under light microscopy. Cell culture medium was refreshed every two days and passages were done 5 days following cell-seeding. Cell counts and viability determination was converted with an automatic cell counter utilizing trypan blue exclusion method. Xenograft and orthotropic mouse model was established by injecting OSC-19 cells to the tongues of nude mice, and tumor progression was monitored. Results: OSC-19 cell line was monitored to have invasive proliferation following 8 passages in a month, and became confluent in 5 days following a passage. Cells were determined to have (>93%) viability before transplantation to mice. For establishment of orthotrophic mouse model (n=2) nude mice were utilized, where (1x106) OSC-19 cells were determined as ideal number to be injected within 25µL of cell culture medium. Tumor development was observed on day 8 post injection, and tumor growth sufficient to block nutrition and respiration was achieved on day 24. Conclusions: In light of these findings, xenograft oral squamous cell carcinoma mouse model has been established. This manuscript defines the key points of these experiments and difficulties faced, all of which are expected to be helpful for future studies. This experimental model is expected to be beneficial in prospective studies that aim to investigate current or novel therapeutic approaches.
