Abstract

Aim: Despite its antineoplastic effect, doxorubicin (DOX) causes significant oxidative stress and cellular damage in the liver. This study aimed to demonstrate the oxidative damage induced by DOX in liver tissue at the biochemical, histological, and molecular levels and to evaluate the protective effects of Montelukast (ML) on this damage. Material Method: To further evaluate the oxidative stress induced by DOX in liver tissue, superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) activities, reduced glutathione (GSH) levels, and malondialdehyde (MDA) levels were analyzed (p<0.05). Results: Histopathologically, portal congestion, pyknotic nuclei in hepatocytes, cellular degeneration, and sinusoidal dilation were observed. In the DOX + ML group, oxidative stress markers improved, SOD, CAT, GPx, and GSH decreased, while MDA levels increased, histological findings were attenuated, and apoptotic gene expression (Bax, Bcl-2, Caspase-3) was significantly modulated compared to the DOX group. Conclusion: The findings suggest that ML provides a significant hepatoprotective effect against DOX-induced oxidative liver damage.

Özet

Amaç: Doksorubisin (DOX), antineoplastik etkisine rağmen karaciğerde belirgin oksidatif stres ve hücresel hasara yol açmaktadır. Çalışmada DOX’un karaciğer dokusunda oluşturduğu oksidatif hasarın biyokimyasal, histolojik ve moleküler düzeyde ortaya konulması ve Montelukast’ın (ML) bu hasar üzerindeki koruyucu etkilerinin değerlendirilmesi amaçlanmıştır. Material Metot: DOX uygulamasının karaciğer dokusunda oluşturduğu oksidatif stresin daha kapsamlı değerlendirilmesi amacıyla Süperoksit Dismutaz (SOD), Katalaz (CAT), Glutatyon Peroksidaz (GPx) aktiviteleri ile indirgenmiş glutatyon (GSH) düzeyleri ve malondialdehit (MDA) seviyeleri analiz edilmiştir (p<0.05). Bulgular: Histopatolojik olarak portal alanlarda konjesyon, hepatositlerde piknotik nükleus, hücresel dejenerasyon ve sinüzoidal genişleme tespit edilmiştir. DOX + ML grubunda oksidatif stres göstergeleri iyileşmiş, SOD, CAT, GPx ve GSH azalırken MDA seviyeleri artış göstermiştir, histolojik bulgular hafiflemiş ve apoptotik gen ekspresyonları (Bax, Bcl-2, Caspase-3) DOX grubuna kıyasla anlamlı düzeyde modüle edilmiştir. Sonuç: Bulgular, ML’nin DOX’a bağlı oksidatif karaciğer hasarına karşı belirgin hepatoprotektif etki sağladığını göstermektedir.
