FARE OVUMLARININ İN VİTRO FERTİLİZASYONU ÜZERİNDE ÇALIŞMALAR

Özet

Bu çalışmada deney hayvanı olarak kullanılan farelerde in vitro fertilizasyon için önerilen yöntemlerin denenınesi ve in vitro yolla fertilizasyon olgusunun gerçekleştirilmesi amaç- lanmıştır. Çalışmada ovum elde etmek amacıyla seksüel olgunluğa ulaşmış İsviçre albinosudişi farelere 48 saat ara ile önce PMSG daha sonra HCG hormonu enjekte edilerek süperovulasyon oluşturuldu. Ovumlar, HCG enjeksiyonundan 16 saat sonra fareler öldürilierek cumulus oophorus hücrP.lert ile birlikte elde edildi ve sıvı paratin ile kaplanm_lş, ortasında 0.4 ml vasat bulunan petri kabındaki vasat içerisine kondu. Sperinatozoitler ise İsviçre albinosu olgun erkek farelelin cauda epididimislerinden sağlandı ve 37°C'dek! '?';) 5 co2 ·H etüvde 1.5-2 saat süreyle tnkübe edildi. !nkübasyon vasatı olarak glukoz, sodyum ptruvat, sığır serum albumini ve antibiyotikleri ihtiva eden TiiY vasatı kullanıldı. Daha sonra önceden hazırlanmış bulunan spermatozoit süspansiyonundan ı 0-20 J.L]. bir pipet yardımıyla alınarak ovumlann bulunduğu petri kabına ilave edildi ve 6 saat süreyle birarada inkübasyona bırakıldılar. Bunu takiben ilk deneme grubundaki fertilize ovumlar aynı vasat içerisinde bırakılmalanna karşın ikinci deneme grubundaki fertilize ovumlar gelişme vasatı olan, ancak sodyum laktat içermeyen Modifiye Whitten's vasatma nakledildi ve 37°C'deki % 5 C02'li etüvde inkübasyonu takiben mikroskop altında kontrol edildiler. Sonuç olarak Ovum ve spermatozoitlerin birarada inkübasyonlan sonucu ovumlann fertilizasyonlan sağlanmış ancak daha ileri bölünme aşarnalanna kadar geliştirilememiştir.

Abstract

The aim of this study was to test the recommended techniques of in vitro fertilization in Iriice as an expeıimental animaL Adult female mice of Swiss albino strain were superovulated by intrapeıitonal injection of PMSG and HCG 48 hours apart. They were killed 16 hours after injection of HCG and their eggs were recovered under warm oil in a plastic dish containing 0.4 ml of ineubatian medium. Sperms recovered from cauda epididymis of adult males of the same strairi were suspended in a medtum and incubated at 37° C under % 5 co2 in air for 1.5-2 hours before they were used for insemination. The ineubatian medium was TifY medium containing glucose, sodium pyruvate, bovine albumtn and antibiotics. At the time of insemination, a drop of sperm suspension (10-20 J.L].) was added to the medium contatning egg clot. Eggs and sperms were incubated in ineubatian medium for 6 hours. In the fırst group, fertilized eggs were left into the ~ame medium. In the second group they were transferred to the development medium (Modifıed Whitten's) without sodium lactate and incubated until they were examtned under stereo-microscope. As a conclusion, in this study fertilized eggs and 2-cell stage embıyos were obtained but further stage of embıyos {morulae, blastocyst) couldn't be reached.
