Özet

Kemoterapide, kanser hücrelerini öldürmek için sitotoksik antineoplastik ajanlar başroldedir. Antikonsülvan kullanan hastalar antineoplastik bir ajanı da kullanmayı gerektiren kanser tedavi sürecine girebilmektedir. Antineoplastiklerin toksik etkisinin, birlikte uzun süreli kullanılan antiepileptiklerle daha çok arttığı bilinmektedir. Bilimsel ilerlemeler ve eczacılık tekniklerinin gelişmesiyle bitkilerin tedavi edici değere sahip etken maddeleri kanser gibi birçok hastalığın tedavisinde kullanılmaya başlanılmıştır. Bu çalışmada, sisplatin ve valproik asitin uzun süreli kullanımlarının meyve sineklerinin yaşama yüzdesi ve ömür uzunlukları üzerindeki olası toksik etkinin kudret narı ile giderilip giderilmeyeceği araştırılmıştır. Bu amaçla, kontrol, 0.05mM valproik asit, 0.05mM sisplatin ile 2.5 ve 5mg/mL kudret narının çekirdek ve meyvelerine ait metanol ekstrelerini içeren deney setleri hazırlanmıştır. Ömür uzunluğu deneyleri, Drosophila melanogaster‘in Oregon R soyuna ait dişi ve erkek populasyonlarında ayrı ayrı çalışılmıştır. Kontrol ve uygulama gruplarına ait sonuçların istatistiksel analizleri için tek yönlü varyans analizi uygulanmıştır. Çalışmamızda valproik asit ve sisplatine maruz kalan hem dişi hem de erkek populasyonlarda ömür uzunluğu ve yaşama yüzdesi bakımından kontrole göre önemli bir düşüş olduğu belirlenmiştir. Kudret narının çekirdek ve meyve ekstrelerinin valproik asit ve sisplatinle birlikte uygulandığı deney gruplarında yaşama yüzdesi ve ömür uzunluğu değerlerinin kontrole yaklaştığı gözlenmiştir. Sisplatin+valproik asit grubunda daha fazla olduğu gözlenen toksik etkiye karşı kudret narının koruyucu etkisinin, bitkinin sahip olduğu bileşenlerin yüksek antioksidan aktivitelerinden kaynaklandığı düşünülmektedir.

Abstract

In chemotherapy, cytotoxic antineoplastic agents play a leading role in killing cancer cells. Patients using anticonvulsants may also undergo cancer treatment which requires the use of an antineoplastic agent. It is known that the toxic effect of antineoplastics increases during a long time with using antiepileptics. With the advancement of scientific advances and the development of pharmaceutical techniques, the therapeutic agents of plants have been used in the treatment of many diseases such as cancer. In this study, it was investigated whether the long-term use of cisplatin and valproic acid can be eliminated with bitter melon by the possible toxic effect on the survival rate and lifespan of fruit flies. For this purpose, control and application sets containing 0.05mM valproic acid and 0.05mM cisplatin and methanol extracts of 2.5 and 5mg/mL bitter melon were prepared. Lifespan experiments were separately studied in male and female populations of the Oregon R strain of Drosophila melanogaster. One-way analysis of variance was used for statistical analysis of control and application groups. In our study, a significant decrease was found in both female and male populations exposed to valproic acid and cisplatin in terms of lifespan and survival rates compared to the control. It was observed that the survival percentage and lifespan closed to the control group in the experimental groups where bitter melon extracts with valproic acid and cisplatin. It is thought that the protective effect of bitter melon against the toxic effect observed in cisplatin+valproic acid group is due to the high antioxidant activities of the plant components.
