Determination of Bean Common Mosaik Virus in Bean Growing Areas of Burdur Province by Serological and Molecular Methods

Özet

Bu çalışma, Burdur ili fasulye yetiştiriciliği yapılan alanlarda Fasulye adi mozayik virüsü (Bean common mosaic virüs: BCMV)’ünün belirlenmesi amacıyla gerçekleştirilmiştir. Sürvey çalışmalarında virüs simptomu gösteren bitkilerden 102 yaprak örneği alınmıştır. Örneklere Double Antibody Sandwich Enzyme Linked Immunosorbent Assay (DAS-ELISA) ve Immunocapture Reverse Transcriptase-Polimerase Chain Reaction (IC-RT-PCR) testleri uygulanarak Bean common mosaic virüsü (BCMV) tanılanmıştır. Yürütülen çalışmalar sonucunda toplam 24 yaprak örneğinde (% 23.52) BCMV belirlenmiştir. IC-RT-PCR yöntemi ile enfekteli olan bu 24 örneğin tamamı, DAS-ELISA yöntemi ile ise 18’i saptanmıştır. IC-RT-PCR yönteminde spesifik primerler ile kılıf protein geninin yaklaşık 850 bp’lik bir kısmı amplifiye edilmiş ve BCMV’ne özgü beklenen seviyede bant elde edilmiştir.

Abstract

The aim of this study was to determine the presence of Bean common mosaic virus; BCMV of bean plants grown in Burdur province. Throughout surveys, a total of 102 leaf samples was collected from plants showing disease symptoms. By employing Double Antibody Sandwich Enzyme Linked Immunosorbent Assay (DAS-ELISA) and Immunocapture Reverse Transcriptase-Polimerase Chain Reaction (IC-RT-PCR) methods, Bean common mosaic virus was identified. Based on research results, BCMV have been found in 24 samples (23.52 %) and all of the 24 infected samples were detected IC-RT-PCR methods. Besides as a results of DAS-ELISA tests it was observed that 18 samples out of 102 were infected with BCMV. IC-RT-PCR was carried out by using spesific primer which amplified a 850 bp fragment of coat protein of BCMV in samples. Thus, BCMV product of expected size in leaf samples were observed.
