Leiomyom ve normal myometriumda immunohistokimyasal vasküler endoteliyal growth faktör (VEGF) ekspresyonu

Özet
Amaç: Myoma uteri nedeniyle histerektomi uygulanan olguların normal myometrium düz kas hücrelerinde ve leiomyoma dokusunda immunohistokimyasal olarak VEGF ekspresyonları karşılaştırılarak VEGF’in myoma uteri gelişimindeki önemi araştırılmıştır. Materyal ve Metod: Myoma uteri nedeniyle histerektomi uygulanan perimenopozal dönemdeki 20 olgu çalışma kapsamına alındı. Olgulardan elde edilen histerektomi materyalinde normal myometrium dokusu ve leiomyomlarda immünohistokimyasal boyama yapılarak VEGF histolojik skorlaması bakıldı. Kesitler her yoğunluktaki boyama için hücrelerin yoğunluğunun ve yüzdesinin ayrı ayrı hesaplandığı semi-kantitatif bir yolla değerlendirilmiştir. Boyanma yoğunlukları; 0 (boyanmamış), +1 (az boyanmış), +2 (belirgin boyanmış) ve +3 (çok kuvvetli boyanmış) olarak sınıflandırılmış ve her grubun %10’u pozitif boyanmış hücre yüzdesini hesaplamak için kullanılmıştır. Her lam için histolojik değer hesaplanarak elde edilen sonuçlar karşılaştırıldı. histolojik değer= Toplam (yoğunluk x hücre yüzdesi) (HSCORE= E (I x PC) ). Bulgular: Leiomyom ve myometriumda elde edilen ortalama VEGF skorları karşılaştırıldığında myometrium grubunda elde edilen skor daha yüksekti ve aradaki fark istatistiksel olarak anlamlı idi. Abstract
Objective: The aim of the study was to determine immunohistochemical expression of VEGF in leiomyomas and normal myometrium in women who histerectomized due to myoma uteri. Materials and Methods: Twenty women who had undergone histerectomy due to myoma uteri were included to the study. Histologic VEGF scorings of the leiomyomas and normal myometrial tissue samples detected with immunohistochemical staining. All sections were scored in a semiquantitative fashion, which considers both the intensity and percentage of cells staining at each intensity. Intensities were classified as 0 (no staining), +1 (weak staining), +2 (distinct staining), and +3 (very strong staining), whereas 10% groupings were used for the percentage of cells that stained positive. For each slide, a value designated histologic score was obtained and the results were compared. HSCORE = (I x PC), where I and PC represent intensity and percentage cells that stain at each intensity.
