Production of anti-human immunoglobulin G ( Anti-HIgG ) antibodies and ıts application in the detection of hepatitis C virus ( HCV ) antibodies with avidin-biotin solid phase enzyme immunoassay ( EIA )

Özet

Hepatit C virus antikorları için bir avidin-biotin solit faz enzim immunoassay (EIA) tanımlanmıştır. Human immunoglobulin G (HlgG) tavşanları immunize etmede kullanılmıştır. Poliklonal antikorlar sepharose A kullanılarak pürifiye edilmiş peroksidaz suksinimit biotin esteri kullanılarak biotinlenmiştir. Probe detektör reagent olarak streptavidin konjuge horseradish peroksidaz (HRP) kullanılmıştır. Üretilen El A, piyasadan satın alınan anti HlgG-HRP konjugatı (VBI HCV EIA, Organon Teknika, Hollanda) ile daha önceden doğrulanmış 15 pozitif ve 25 negatif serum örneği kullanılarak karşılaştırmıştır. Sonuçlar, elde edilen konjugatın anti-HCV IgG saptanması açısından, ticari konjugatla eşdeğer sonuçlar verdiğini göstermiştir. Bu çalışma, Türkiye'de yapılan ilk anti HCV-EIA konjugat üretimidir.

Abstract

An avidin-biotin solid phase enzyme immunoassay (EIA) for Hepatitis C Virus (HCV) antibodies was described. Human immunogloblin G (HlgG) was used for the immunization of rabbits. Resulting polyclonal antibodies were purified by HlgG mobilized. Sepharose A and biotinylated peroxidase using a succinimide ester of biotin. Streptavidin-conjugated horseradish peroxidase (HRP) was used as a probe detector reagent. Biotinylated rabbit anti-HIgG was compared with commercial goat anil HlgG-HRP conjugate (UBI HCV EIA, Organon Teknika, the Netherlands) using coated plates with synthetic peptides representing epitopes of Hepatitis C Virus (HCV) for the presence of HCV antibodies in 15 positive and 25 negative donor sera. The results showed that the conjugate is of equivalent detectability in showing antibodies in IgG nature raised against HCV. This study has been the first succesful production of in-house conjugate for anti HCV EIA in Turkey.
