Hiperbarik oksijen tedavisinin fötal kortikal greftler üzerine etkisi

Özet
Nöral doku transplantasyonlarında greftin yaşama şansını etkileyebilen bir çok iç ve dış unsur söz konusudur. Transplantasyon sonrasında greftin konakçı ile anatomik bir bütünlük oluşturabilmesi için belli bir süreye gereksinimi vardır. Henüz damarlanması tamamlanmadığı için de greftte en yüksek oranda hücre kaybı bu dönemde olmaktadır. Bu çalışmada transplantasyon sonrası deneklere hiperbarik oksijen tedavisi (HBOT) uygulayarak bu tedavinin greftin gelişimi ve konakçıyla bütünleşmesi üzerine etkilerini araştırdık. Çalışmamızda 30 adet erişkin Wistar albino sıçan kullandık. Sıçanlar her bir grupta 10 adet denek olacak şekilde 3 gruba ayrıldı. Birinci grupta sadece frontal loba boşluk açıldı ve HBOT uygulandı. İkinci grupta lobda yer açılmasını takiben 16 veya 17 günlük fötal sıçan kortikal dokuları transplante edildi. Üçüncü grupta ise aynı şekilde transplantasyonu takiben HBOT uygulandı. HBOT'ne transplantasyon sonrası 1.saatte başlanıldı ve 5 gün devam edildi. Sıçanlara günde iki kez 90 dakika süreyle 2 ATA (atmosphere absolute) basınçlı % 100 oksijen uygulandı. Sıçanlar, ameliyat sonrası 7. günde radyolojik incelemelerini takiben feda edildiler. Radyolojik ve histolojik değerlendirmelerde greftlerin yaşama ve büyüme oranlarının üçüncü grupta daha yüksek olduğu, konakçıyla bütünleşmelerinin yeterli düzeye ulaştığı saptandı.
Abstract
Both extrinsic and intrinsic factors affect the survival of neural tissue transplantation grafts. Grafts needs a time period for integration with the host after the transplantation. Cell loss of the graft is highest in this period since vascularization has not been completed yet. In our study, we investigated the effect of hyperbaric oxygen treatment (HBOT) on development rate and integration of the grafts. We used 30 Wistar albino rats. Rats were divided into three groups of 10 rats. In the first group only cavitation in the frontal was performed and then HBOT was given. In the second group, we transplanted fetal cortical tissue which was obtained from rat fetuses of 16-17 days of age following cavitation. In the third group we applied HBOT after the same transplantation procedure. HBOT was begun at the first hour after transplantation and was continued for 5 days. 100 % oxygen in 2 absolute atmosphere (ATA) pressure was applied to the rats for 90 minutes in each session, twice a day.Rats were sacrified at the end of the 7th day, after radiological evaluation. Analysis of histological and radiological findings showed that in addition to survival and development rate, the degree of integration of the grafts reached to optimum level in the third group.
