Özet

Bu araştırmada beyaz Leghorin tipi tavuk embriyolarında, nöral tüp kapanma defekti gelişimi üzerine etanolun etkisi ışık mikroskopu altında histopatolojik olarak araştırıldı. Çalışmamızda 40 adet (n: 40), spesifik patojen içermeyen (SPF), fertil, beyaz Leghorin tipi tavuk yumurtası iki eşit gruba ayrıldı (n: 20). Her iki grup 37, 8°C ± 0,2°C'da 30 saat inkübe edilerek embriyonel gelişimlerinin 8. basamağına ulaştırıldılar. Birinci grup kontrol grubunu oluşturdu. İkinci grup çalışma grubu olup, in-ovo 42 mg/ dl tek doz etanol verildi. Her iki gruptaki yumurtalar 48. ve 72. saatler sonunda açılarak ışık mikroskopu altında histopatolojik olarak değerlendirildi. 48. ve 72. saatlerde açılan yumurtalarda, kontrol grubunun tamamında normal embriyolojik gelişimin devam ettiği, plasental damarlanmanın oluştuğu, kalp aktivitesinin başladığı ve nöral tüpün kapandığı gözlendi. Ancak çalışma grubundaki embriyoların kontrol grubundakilere göre belirgin gelişme geriliği gösterdikleri, mikrosefalik oldukları ve nöral tüp kapanma defekti oluştuğu gözlendi.

Abstract

In this study, the effects of ethanol on the formation of the neural tube closure defects in White Leghorn chick embryos were investigated under the light microscope. Fertile specific pathogen free (SPF), White Leghorn eggs (n:40) were divided into two groups (n: 20). Both of them were incubated at 37.8°C ± 0.2°C and 65-75 % relative humidity during 30 hours and the eighth stage of the embryonic development was reached (Hamburger and Hamilton). The first group was control group (n: 20). The second was the study group and it was given in-ovo a single dose, 42 mgr/100 cc of ethanol. The eggs in both groups were opened and investigated under the light microscope at the end of the 48th and 72 th hours. The embryos in the control group were developed normally according to Hamburger and Hamilton classification. The normal placental vessel development, cardiac activity and closure of the neural tube were seen. However in the study group, microcephaly, neural tube closure defect and significant developmental retardation were noticed as compared to the control group.
