Bir kaynak suyu tesisinde olası mikrobiyal kontaminasyonun incelenmesi

Özet
Bu araştırmada, bir kaynak suyu tesisinde, kaynaktan son ürüne kadar, çeşitli noktalardan numuneler alınarak hangi aşamaların mikrobiyal risk taşıdığı incelenmiştir. Ayrıca geri dönüşümlü damacanalar ile bunların dezenfeksiyon ve durulamalarında kullanılan sular da kontrole tabi tutulmuştur. Alınan su örnekleri, 2005 tarihli yönetmelikte bildirilen toplam bakteri sayısı, koliform bakteri sayısı ve Escherichia coli yönünden incelenmiştir. Toplam mezofilik aerop bakteri sayısı bakımından kaynağa ait su örneklerinin % 65'i, diskli membran filtreden alınan su örneklerinin % 60'ı, ters osmoz cihazından alınan su örneklerinin % 90'ı, temiz su toplama havuzundan alınan su örneklerinin % 10'u, dolum musluklarından alınan su örneklerinin % 60'ı, dolum yapılmış damacanalardan alınan su örneklerinin % 50'sinin ilgili yönetmeliğe uygun olduğu bulunmuştur. Kaynaktan, diskli membran filtreden, ters osmoz cihazından, temiz su toplama havuzundan alınan örneklerde koliform bakteri ve Escherichia coli tespit edilmiştir. Dolum musluklarından alınan örneklerin % 15'i, dolum yapılmış damacanalardan alınan örneklerin ise % 10'unun koliform bakteri yönünden yönetmeliğe uygun olmadığı belirlenmiştir. Geri dönüşümlü polikarbon damacanalar ile bunların dezenfeksiyon ve durulama sularında toplam bakteri ve koliform bakteri içerdiği saptanmıştır. Örneklerin hiç birinde Escherichia coli tespit edilmiştir. İşletmedeki en önemli kontaminasyon kaynağının kirli gelen ve iyi temizlenip dezenfekte edilemeyen dönüşümlü polikarbon damacanalar olduğu saptanmıştır.
Abstract
In this study, microbiological hazard points of water were investigated from the start point of spring to the drinkable final product, was performed in a spring water filling plant. On the other hand, demijohns and the washing water of the demijohns were also analized. The total bacteria counts, coliform and E. coli analises were applied to the water samples according to 2005 dated regulation. The total bacteria counts in 65 % of the samples collected from spring, 60% of the samples collected from discus membrane filter, 90 % of the samples collected from reverse osmosis system, 10 % of the samples collected from clean water keeping pools, 60 % of the samples collected from filling taps and 50 % of the samples collected from filled demijohns found to be acceptable according to the values given in the regulation. No coliform and Escherichia coli were detected in the samples collected from spring, discus membrane filter, reverse osmosis system, clean water keeping pools. Coliform counts were over the limit values given in the regulation in 15 % and 10 % of the samples collected from filling taps and filled demijohns, respectively. Escherichia coli was detected from none of the collected samples.It was concluded that the most important contamination source in the plant was insufficient cleaned and disinfected dirty polycarbon demijohns.
