The possible role of nitric oxide in the evasion of the immune system by Toxoplasma gondii: Serum nitrite levels in acute and chronic phases of toxoplasmosis in humans

Özet
Toxoplasma gondii insan vücudunda iki şekilde bulunur: takizoitler ve bradizoitler. Takizoitler infektif formu, bradizoitler ise bağışıklık sisteminden saklanma formu olup, insan vücudunda Toxoplasma gondii'nin uzun süre yaşamasına destek olurlar. Nitrik oksit (NO), insanların özgün olmayan bağışıklık sisteminin bir bölümüdür ve birçok deney NO' nün anti toksoplazmik rolünü göstermektedir. Toxoplasma gondii' nin bağışıklık sisteminden saklanışı in vitro deneylerde gösterilmiş olmasına karşın, in vivo olarak henüz gösterilmemiştir. Bu çalışmada gebe olmayan (n= 107) olguların serumları Toxoplasma IgM ve IgG EIA (Organon Technica Inc., Holland) ile çalışıldıktan sonra, nitrik oksidin in vivo saklanma dönemindeki olası rolünü saptamak üzere, Griess Reagent ile değerlendirildi. Grupların serum nitrit değerleri : IgM -/IgG - kontrol grubunda (n=10): 19.6+0.6 uM (95 % Confidence interval, C.I: 19.25-19.95); IgM -tVIgO - grubunda (n=10): 6.0610.9 uM (C.I: 5.52-6.60); IgM +/ IgG + (15-300 lU/mL) (n=31): 19.0611.91 UM (C.I: 18.4-19.72); IgM -/IgG + (15-150 lU/mL) (r.=34): 38.714.6 uM (C.I: 37.18-40.22); IgM -/IgG + (150-300 lU/mL) (n=32): 13.513.6 uM (C.I: 12.27-14.73) olarak saptandı. Abstract
Toxoplasma gondii can survive in human body in two forms: tachyzoites and bradyzoites. Tachyzoites are the infective forms but bradyzoites are the forms which evade the immune system and which support the long time survival of Toxoplasma gondii in human body. Nitric oxide (NO) takes part in human non-specific immunity and many experiments have shown its anti-toxoplasmic role. The evasion from the immune system by Toxoplasma gondii has been shown using in vitro experiments, but not by in vivo experiments. In this study, after the sera of non pregnant women (n=I07) were examined by Toxoplasma IgM and IgG EIA, we examined their serum nitrite levels using the Griess Reagent (Organon Technica Inc., Holland) in order to determine the possible in vivo role of nitric oxide in the evasion period. The serum nitrite level in the IgM -/IgG -control group (n=10) was 19.6+0.6 uM (95 % Confidence interval (C.I): 19.25-19.95). In the IgM +/IgG - group (n=10) it was 6.06±0.9 uM (C.I: 5.52-6.60). to the IgM +/ IgG + group (15-300 lU/ml) (n=31) it was 19.06+1.91 uM (C.I: 18.4-19.72). In IgM -/IgG + (15-150 lU/mL) group (n=34) it was 38.7+4.6 uM (C.I: 37.18-40.22). In the IgM -/IgG + (150-300 lU/mL) group (n=32) it was 13.5+3.6 uM (C.I: 12.27-14.73).
