Özet

Fasciola hepatica sıklıkla sığır ve koyunlarda nadiren de insanlarda fasciolosise neden olan bir karaciğer trematodudur. Fasciolosis dünyada ve ülkemizde yaygın olarak bulunmakta ve özellikle hayvancılık sektöründe önemli ekonomik kayıplara yol açmaktadır. F. hepatica'nm ES antijenlerinden olan Cathepsin Ll parazitin dokulara penetrasyonda, immun yanıttan kaçmasında ve beslenmesinde önemli rolü olan bir enzimdir. Özgül antijenik yapısı nedeniyle tanı yöntemlerinde ve korunma amacıyla immünizasyon çalışmalarında kullanılabilecek potansiyel bir moleküldür. Bu çalışmada F. hepatica cathepsin Ll geninin klonlanması ve okaryotik hücrelerde belirlenmesi amaçlanmıştır. Bu amaçla erişkin F. hepatica'dan toplam RNA izolasyonu yapılarak tersine transkripsiyon-polimeraz zincir reaksiyonu (TT-PZR) ile cathepsin Ll DNA amplikonları oluşturuldu. Cathepsin Ll genine özgü primerler kullanılarak 981 bazlık cathepsin Ll geni kodlama bölgesi çoğaltıldı. Daha sonra, cathepsin Ll geni pCI-neo açıklama vektörüne klonlanarak PZR-T ve enzim kesim deneyleri ile genin varlığı doğrulandı. Oluşturulan rekombinant plazmit, pFhCLl olarak adlandırıldı. Rekombinant pFhCLl plazmiti Vero hücrelerine geçici olarak transfekte edilerek cathepsin Ll proteinin varlığı Western immuno blot tekniği ile gösterildi.

Abstract

The parasitic trematode Fasciola hepatica is the causative agent of fasciolosis that is common in ruminants especially sheep and cattle and is occasionally found in humans. Fasciolosis has a worldwide distribution including Turkey and causes major economic losses in agricultural industry. Cathepsin Ll is one of the major molecules in the excretory-secretory products of F. hepatica and is involved in tissue penetration, immune evasion and feeding and therefore may be used in vaccination and serological diagnosis. The aim of this study was to evaluate cloning and expression of the cathepsin Ll gene of F. hepatica eucaryotic cells. For this purpose, total RNA was extracted from adult F. hepatica. Cathepsin Ll DNA amplicons were obtained with the reverse transcription polymerase chain reac¬tion (RT-PCR). The 981 base-coding gene region of cathepsin Ll was amplified using specific primers to the cathepsin Ll gene. Then, the cathepsin Ll gene was cloned into the pCI-neo mammalian expression vector. The presence of the cathepsin Ll gene was confirmed by PCR screening and enzyme digestion assays. So, the resulting recombinant plasmid was named pFhCLl. Afterwards, the pFhCLl vector was transiently transfected into Vero cells. The presence of the cathepsin Ll proteins was shown by Western immunoblotting.
