Aydın yöresindeki köpeklerde echinococcus granulosus yaygınlığının polimeraz zincir reaksiyonu ile belirlenmesi

Özet

Amaç: Çalışma Aydın ve yöresindeki sahipli köpeklerde E. granulosus yaygınlığını doymuş tuzlu su flotasyon ve PCR yöntemleriyle belirlemek amacı ile yapılmıştır. Yöntemler: Çalışmada kullanılacak materyal, Aydın ve yöresinde bulunan, 6 aydan büyük, sahipli 100 köpekten taze ve temiz olmak koşuluyla yerden toplanmıştır. Örnekler Aydın merkez ve 5 ilçe (Söke, Koçarlı, Çine, Bozdoğan, Kuyucak) ile birlikte 6 bölge olmak üzere toplam 13 köyden alınmıştır. Dışkı örneği alınan köpeklerin 25’i dişi, 75’i erkek olup yaşları 6 ay-16 yaş aralığındadır. Dışkılar öncelikle makroskopik ve tuzlu su flotasyon yöntemi ile incelenmiş daha sonra E. granulosus varlığı PCR yöntemiyle araştırılmıştır. Bulgular: Bir köpekte (%1) E. granulosus’a rastlanmıştır. Ayrıca incelenen köpek dışkılarının 11’inde (%11) Toxocara canis, 3’ünde (%3) Ancylostoma caninum, 2’sinde (%2) Taenia spp. ve 1’inde (%1) Capillaria spp. yumurtalarına rastlanılmıştır. Sonuç: Toplam 100 hayvandan 1’inde E. granulosus varlığı Türkiye’nin diğer bölgelerinde yapılan çalışmaların çoğuna göre düşük bulunmuştur. Gerek makroskopik gerekse mikroskobik bakıda parazitolojik yönden negatif bir dışkıda E. granulosus tespit edilmesi hayvan sahipleri, veteriner hekimler ve parazitoloji laboratuvarı çalışanları için köpek dışkılarının her zaman potansiyel tehlike oluşturduğu gerçeğinin altını çizmektedir.

Abstract

Objective: In this study, it was aimed to determine the prevalence of E. granulosus in the Aydın district. Methods: Clean and fresh faeces samples were collected off the ground from owned dogs (n=100; 25 female, 75 male) with the range of 6-16 months old. These faeces samples were obtained in 13 villages from five different locations (Centre, Söke, Çine, Bozdoğan and Kuyucak) of the Aydın district. The prevalence of E. granulosus was investigated with PCR and prior to applications of molecular techniques, faeces samples were investigated with macroscopic and floatation methods Results: E. granulosus was determined in only one dog (1%; 1/100) and Toxocara canis (11%), Ancylostoma caninum (%3), Taenia spp. (2%) and Capillaria spp. (1%) eggs were also identified. Conclusion: Current prevalence (1%) of E. granulosus in this study is lower than the other studies conducted in different parts of Turkey. Determination of E. granulosus with PCR in faeces without determining any parasites with either macroscopic or microscopic examinations could indicate the potential risk of dog faeces for animal owners, veterinarians and parasitology laboratory technicians.
