The effect of human granulocyte and granulocyte macrophage-colony stimulating factors on neutrophil chemiluminescence

Özet

Granülosit ve Granülosit-Makrofaj Koloni Uyarıcı Faktörlerin Nötrofil Kemiluminesansına Etkileri Kanserli hastaların tedavilerinde en önemli mortalite nedenlerinden birisi nötropeniye sekonder gelişen enfeksiyonlardır. Son yıllarda geliştirilen granülosit ve granülosit-makrofaj koloni uyarıcı faktörler (G-CSF), '(GM-CSF) nötropeni süresini kısaltarak ve/veya derin nötropeniye girişi önleyerek kanser morta-lite ve morbiditesini azaltan ve daha etkili tedavi metodlannm kullanımım olanaklı kılan sitokinlerdir.dir.Özellikle myeloablatif tedaviler sonrası yapılan kök hücre nakilleri tecrübelerinde doğal öldürücü hücrelerde gösterildiği üzere bir immünişlemci hücre popülasyonu sayısının normale gelmesi o fonksi-yonun normale gelmesi anlamında olmamaktadır. Nötropeninin sayı olarak düzelmesinin fonksiyonel düzelmeye eşlik edip etmediğinin hızlı bir şekilde değerlendirilebilmesi amacıyla luminol bağımlı kemi-luminesans testi geliştirildi ve bununla G-CSF ve GM-CSF 'in nötrofil oksidatif patlamasına olan etkisi incelendi. Rekombinant iman G-CSF optimal olarak 180-240 ng/mL, GM-CSF ise 120-180 ng/mL doz-larında doz bağımlı bir şekilde nötrofil oksidatif patlamasını arttırdılar. Opsonize zimozan ile inkübe edilmiş nötrofillerin oksidatif patlamasını ise G-CSF arttırırken GM-CSF arttırmadı. Zimozan CR3 (kompleman reseptör 3) ile etkileşmektedir. Bu sonuçlar daha kök hücrelere yakın öncü hücrelere etki eden GM-CSF'in hücresel ve humoral immün sistem etkileşiminin bir parçası olarak kompleman resep-törü üzerinden sinerjizminin olmadığını, G-CSF'in ise olabileceğini düşündürmüştür.

Abstract

One of the most important causes of mortality in patients treated for cancer is secondary infections re-lated to neutropenia, Granulocyte-colony stimulating factor (G-CSF) and granulocyte macrophage-co-lony stimulating factor (GM-CSF) are cytokines purified and produced in the past few years. They either prevent neutropenia or shorten the neutropenic phase which decreases cancer mortality and morbidity. Consequently, these factors permit clinicians to use more effective therapeutic medications. The norma-lization of function in an immune effector cell subset does not necessarily overlap with the normaliza-tion of the cell number as observed in natural killer activity after bone marrow transplantation. A lumi-nol-dependent Chemiluminescence assay was developed to evaluate ivhether the repair in neutropenia accompanies the neutrophil ability to function. The effect of G-CSF and GM-CSF on neutrophil respira-tory burst was examined.The optimal enhancing effect of recombinant human G-CSF and GM-CSF on neutrophil oxidative burst was dose dependent, obtained at a 180-240 ng/mL and 120-180 ng/mL dose interval, respectively. The preincubatton of cells with G-CSF enhanced the neutrophil oxidative burst to zymosan activated sera (ZAS), in contrast preincubatton with GM-CSF could not. ZAS interacts with type 3 complement receptors (CR3). These results suggest that GM-CSF which acts on progenitor cells more clo-se to stem cells does not have any synergistic activity with CR3, while G-CSF might be related in such synergism. Thus, G-CSF might be more active in evoking synergism between humoral and cellular im-mune system, at the neutrophil level in cancer patients.
