Özet

Bu çalışmada, depo kanlarına melatonin ilave edilerek kalitelerinin artırılması amaçlandı. Bu amaçla depolanma süresince eritrositlerdeki lipid peroksidasyonu ve eritrosit antioksidan potansiyelindeki değişiklikler ile melatoninin koruyucu etkileri araştırıldı. Beşi kontrol grubu, beşi çalışma grubu l ve beşi çalışma grubu 2'de olmak üzere toplam on beş donörden tam kan alındı. Çalışma grubu 1'in kan torbalarına düşük doz (50 pg/ml), çalışma grubu 2'nin kan torbalarına ise yüksek doz (5000 pg/ml) melatonin eklendi. Kan torbalarından eritrositler (Red Blood Cells - RBC) sayıldı. Hemoliz derecesini değerlendirmek amacıyla plazma potasyum düzeyi ve laktat dehidrogenaz. (LDH) aktivitesi ölçüldü. Eritrositlerdeki lipid peroksidasyonunu değerlendirmek için eritrositlerdeki tiobarbitürik asitle reaksiyona giren substratların (TBARS) düzeyi araştırıldı. Antioksidan sistem hakkında bilgi almak amacıyla eritrosit antioksidan potansiyel (AOP) düzeyleri çalışıldı. Ölçümler, depolanmanın 0. 1, 3., 5., 7., 10., 14., 21., 28. ve 35. günlerinde yapıldı. Abstract

The aim of this study was to improve the quality of stored blood by adding melatonin to the blood bag. Therefore, the changes in lipid peroxidation and antioxidant potential during storage of erythrocytes and protective effect of melatonin were investigated. The whole bloods were taken from fifteen donors, which of five was control group, five was study group 1 and the rest five was study group 2. Low dose (50 pg/ml) and high dose (5000 pg/ml) of melatonin was added to the blood bags of study group 1 and study group 2, respectively. Red Blood Cells (RBC) were counted in whole blood. Potassium level and lactate dehydrogenase (LDH) activity were measured in plasma for determination of hemolysis. The levels of thiobarbituric acid reactive substance (TBARS) and antioxidant defense potential (AOP) were studied in erythrocytes for determination of lipid peroxidation and antioxidant system, respectively. The measurements were performed at the day 0, 1, 3, 5, 7, 10, 14, 21, 28, and 35 of the storage. The levels of TBARS and potassium and the activity of LDH increased depending on storage time whereas AOP and RBC decreased (p
