Prolidazın mutlak aktivitesini değerlendirmede fotometrik enzim aktivitesi ölçüm metodunun optimizasyonu

Özet

Prolidaz, prolin içeren dipeptidlerin (X-Pro) peptid bağını koparan ve kollajen metabolizmasında yer alan önemli bir enzimdir. Prolidaz aktivitesi genellikle prolidaz tarafından açığa çıkarılan prolinin miktarının fotometrik olarak ölçüldüğü yöntemlerle belirlenir. Çalışmada amacımız, bu prolidaz aktivitesi ölçüm yönteminin mutlak aktiviteyi yansıtıp yansıtmadığını araştırmaktır. Yöntemdeki mangan iyonlarıyla yapılan aktivasyon işlemi, proteinlerin çöktürülmesi ve okuma basamakları incelenmiştir. Analiz içi ve analizler arası % varyasyon katsayısı % 10’un üzerinde olan yöntem ile bir test için 6-8 saat zaman harcanmaktadır. Mangan içeren aktive edici reaktif inkübasyon süresince stabil kalmamaktadır ve derişimi enzim aktivasyonu için uygun değildir. Bu nedenlerle, fotometrik yöntemin, mangan derişimleri, ön inkübasyon solüsyonunun pH’sı değiştirilmiş, proteinlerin çöktürüldüğü basamak kaldırılmış ve substratla inkübasyon süresi kısaltılıp ninhidrin tepkimesi pH’sı en uygun hale getirilmiştir. Ortamda var olan ve prolidaz enzimi ile serbestleştirilen prolinin prolidaz enzimini inhibe edebileceği ve yöntemin analitik performansının etkilenebileceği gösterilmiştir. Sonuç olarak prolinin inhibisyonundan en az etkilenecek hale getirilen modifiye (optimize) prolidaz ölçüm metodunun mutlak aktiviteyi daha iyi yansıtabilecek bir yöntem olabileceği düşünülmektedir.

Abstract

Prolidase cleaves the bonds of dipeptides containing proline (X-Pro) and an important enzyme in collagen metabolism. Prolidase activity is generally determined by photometric methods based on the measurement of proline levels produced by prolidase. We aimed to investigate the measured prolidase activity is suitable for the assessment of its physiological activity. Effects of manganese on the enzyme activation, protein precipitation and reading steps of the photometric method, and inhibitory effect of proline on the enzyme were analyzed. The intra- and inter-assay CVs % were higher than 10 % for photometric method and turnaround time was 6-8 hours/test. The activation reagent containing manganese was not stable and its concentration was not optimal for enzyme activation. Thus we modified the photometric method by changing manganese concentrations and pH of activating solution, eliminating protein-precipitating step, arranging the pH of color reagent to the pH optimum for ninhidrin reaction and shortened incubation time with substrate. The inhibitory effect of proline on the prolidase activity even in the physiological and produced proline concentrations during enzymatic analysis may limit the analytical performance of prolidase assays. In conclusion, the modified photometric method presented in this study seems to be more reliable than the classical photometric method and measured prolidase activity may not reflect the true physiological activity of enzyme due to proline inhibition.
