Abstract

Objectives: This study evaluated the association of C677T and A1298C single nucleotide polymorphisms of methylene tetrahydrofolate reductase gene with diabetic retinopathy in type 2 diabetes patients. Materials and Methods: Study subjects comprised of 35 Type 2 diabetes patients. 21 were with diabetic retinopathy, 14 without retinopathy as control subjects. Genotyping of the methylene tetrahydrofolate reductase C677T and A1298C polymorphisms was performed by melting curve analysis of the generated amplicons after real-time online polymerase chain reaction. Serum homocysteine levels were measured by high performance liquid chromatography. Results: The methylene tetrahydrofolate reductase C677T and A1298C genotype allele frequencies were not different between diabetic patients with and without retinopathy (p>0.05). C677T genotype distribution did not differ between control subjects and type 2 diabetes patients with retinopathy (p>0.05). A1298C genotype distribution did not differ between control subjects and type 2 diabetes patients with retinopathy (p>0.05). Serum median homocysteine levels were not significantly different between the two groups (p>0.05). Conclusion: Methylene tetrahydrofolate reductase C677T and A1298C genotype and allele frequencies were not directly associated with the development of diabetic retinopathy in the study group. However further studies with larger scaled groups should be performed in order to investigate the role of C677T and A1298C single nucleotide polymorphisms of methylene tetrahydrofolate reductase gene in type 2 diabetes patients with diabetic retinopathy.
