DFR ve PAL Gen Transkripsiyonu ve Rhodomyrtus tomentosa (Aiton.) Hassk. ‘da Antosiyanin Birikimi ile İlişkisi

Özet
Amaç: Rhodomyrtus tomentosa (Aiton.) Hassk. (R. tomentosa) besin açısından zengindir ve çoklu farmakolojik uygulamalara sahiptir. Antosiyaninler, R. tomentosa’nın çiçek ve meyvelerine renk katmakta ve ışık hasarına karşı koruma sağlamaktadır. Dihidroflavonol 4-redüktaz geni (DFR) ve fenilalanin ammonialyaz geni (PAL) antosiyanin sentezi için çok önemlidir. Gereç ve Yöntem: R. Tomentosa’nın pigmentli organlarında DFR ve PAL transkript seviyeleri ve antosiyanin içeriği sırasıyla qRT-PCR analizi ve spektrofotometri ile incelendi. Gliseraldehit-3-fosfat dehidrogenaz (GAPDH) geni, DFR ve PAL transkript seviyelerinin normalleştirilmesi için referans gen olarak seçilmiştir. Bulgular: DFR ve PAL’nin transkript seviyeleri, güçlü metabolizması olan organlarda, yaşlanan organlardakilere göre daha yüksekti. Meyvenin ilk ve orta olgunluk dönemlerinde DFR ve PAL transkript seviyeleri yukarı düzenlenmiştir. Bu ifade modelleri meyve rengi gelişimi ile tutarlıdır. PAL ve DFR’nin en yüksek transkript seviyeleri orta-olgunluk döneminde veya kırmızı-meyve döneminde gözlenmiştir. Sonuç: R. tomentosa meyvesinin geç olgunlaşma döneminde, iki genin transkript seviyeleri, antosiyaninlerin sürekli olarak birikmesine rağmen, bazı geç olgun meyvelerdeki antosiyanin birikiminden farklı olarak, aşağı düzenlenmiştir.
Abstract
Objectives: Rhodomyrtus tomentosa (Aiton.) Hassk. (R.tomentosa) is rich in nutrients and has multiple pharmacologicalapplications. Anthocyanins confer color to theflowers and berries of R. tomentosa and provide protectionagainst photodamage. The dihydroflavonol 4-reductasegene (DFR) and phenylalanine ammonialyase gene(PAL) are crucial for anthocyanin synthesis.Methods: DFR and PAL transcript levels and anthocyanincontent in the pigmented organs of R. tomentosa wereinvestigated through qRT-PCR analysis and spectrophotometry,respectively. The glyceraldehyde-3-phosphatedehydrogenase (GAPDH) gene was selected as the referencegene for the normalization of DFR and PAL transcript levels.Results: Transcript levels of DFR and PAL were higher inorgans with vigorous metabolism than those in senescentorgans. DFR and PAL transcript levels were up-regulatedduring the initial and middle-maturity periods of fruit.These expression patterns are consistent with fruit colordevelopment. The highest transcript levels of PAL andDFR were observed during the middle-maturity period orthe red-fruit period.Conclusion: During the late maturity period of R. tomentosafruit, the transcript levels of the two genes weredown-regulated even though anthocyanins were continuouslyaccumulated, which was different from the accumulationof anthocyanins in some late mature fruits.
