Özet

Bu araştırmada, taze üretilmiş tulum peyniri iki eşit parçaya bölünmüş ve parçalardan birisi aflatoksin ürettiği bilinen bir Aspergillus parasiticus NRRL 2999 küf suşuyla 104 spor/g düzeyinde aşılanarak, diğeri ise kontrol örneği olarak 10 ± 1 ve 20 ± 1 °C'de % 85 ± 2 ve 95 ± 2 oransal nemde (ON) 90 gün süreyle olgunlaştırılmıştır. Tulum peynirlerinin ambalaj materyali olarak cam kavanozlar kullanılmıştır. Peynir örneklerinde olgunlaştırma süresince 1. günden başlamak üzere 15., 30., 45., 60. ve 90. günlerde ince tabaka kromatografisi (İTK) yöntemi kullanılarak aflatoksin analizi yapılmıştır. Olgunlaştırma süresince 10 ± l °C'de depolanan aşılı ve kontrol grubu örneklerin hiç birisinde tespit edilebilir seviyede aflatoksine rastlanmamıştır. Kontrol grubu örneklerde 20 ± 1 °Ç'de aflatoksine rastlanmazken, aşılı örneklerde 15. günden itibaren aflatoksin oluşumu gözlenmiş ve miktarı zamanla azalmıştır. Doksanıncı gün sonunda % 85 ± 2 oransal nemde 0,98 $mu$ g/kg aflatoksin B,, 47,90 $mu$ g/kg G, ve 4,20 $mu$ g/kg G2; % 95 ± 2 oransal nemde de 32,76 $mu$ g/kg aflatoksin B,, 118,80 ug/kg G, ve 51,98 $mu$ g/kg G2 belirlenmiştir. Aflatoksin B2 ise en son 30. günde % 85 ± 2 oransal nemde 47,25 $mu$ g/kg ve % 95 + 2 oransal nemde 60,50 $mu$ g/kg seviyesinde belirlenmiş ve daha sonraki günlerde saptanamamıştır. Bu araştırmadan elde edilen bulgular, küf bulaşması yanında depolama sıcaklığı ve oransal nem değerlerindeki değişmenin Tulum peynirinde aflatoksin oluşumunu önemli öjçüde etkilediğini göstermiştir. Tulum peynirini 10 °C ve altında depolamanın aflatoksin oluşumu açısından risk oluşturmadığı sonucuna varılmıştır.

Abstract

Tulum cheese was divided into 2 equal parts. Orie part was inoculated with 104 spores/g of aflatoxin-producing AspergiIIus parasiticus NRRL 2999 strain. The other part, the control group, was not inoculated. All the samples were ripened in glass jars at 2 different temperatures (10 ± 1 and 20 ± 1 °C) and relative humidities (85 ± 2% and 95 ± 2%) and analyzed for aflatoxin formation on days 1,15, 30, 45, 60 and 90 of storage using thin layer chromatography. Aflatoxins were determined in the inoculated samples after 15 days with incubation at 20 ± 1 °C, while they were not found in inoculated samples stored at 10 ± 1 °C or in any of the control group samples during a ripening period of 90 days. The levels of aflatoxins decreased after 15 days of storage. In 85 ± 2% and 95 ± 2% relative humidities, the levels of aflatoxins were 0.98 $mu$ g/kg B,, 47.90 $mu$ g/kg G1 and 4.20 $mu$ g/kg. G2; and 32.76 $mu$ g/kg B,, 118.80 $mu$ g/kg G, and 51.98 $mu$ g/kg G2 at the end of 90 days, respectively. The level of aflatoxin B2 was 47.25 $mu$ g/kg and 60.50 pg/kg in 85 ± 2% and 95 ± 2% relative humidities at the end of 30 days, respectively. However, it was not determined on days 45, 60 or 90. Data obtained in this study indicated that storage time and temperature, humidity and aflatoxigenic mould contamination significantly affected aflatoxin formation in Tulum cheese. It was concluded that aflatoxin did not form in Tulum cheese stored at < 10 °C.
