Cation-Chloride Co-Transporters mRNA Expression In Mice CA1 Region After Lithium-Pilocarpine Induced Status Epilepticus

Özet
Amaç: Santral Sinir Sistemi içinde belirli bir hücrenin hücreiçi klorür konsantrasyonu Cl-atıcısistemi ile Cl-biriktirici sistemi arasındaki hassas denge ile oluşturulur. Bu çalışmada lityum- pilokarpin ile oluşturulan status epilepticus fare modelinde katyon-klorür mRNA yardımcı- taşıyıcılarının (CCCs) yapım profili ve CCCs ile epileptogenez arasındaki olası ilişkininaraştırılması amaçlandı.Yöntemler: Balb/c erişkin erkek fareler randomize edilerek iki ayrı gruba ayrıldı: PISE vecontrol grubu olarak. PISE grubundaki fareleri PISE hayvan modelini oluşturmak için üçzaman noktasına ayırdık: 1g, 14g ve 45g. CA1 bölgesinde mRNA CCCs aile üyeleriekspresyonunu ortaya çıkarmak üzere gerçek zamanlı floresan kantitatif PCR (polimerazzincir reaksiyonu) kullanıldı.Sonuçlar: Bu CCCs üyelerinin hepsi kendine özgü bir patern gösterdi. PISEden sonraNKCC1 (sodium potasyum katyon-klorid cotransporter 1), KCC1 (Potasyum katyon-kloridcotransporter 1), KCC3 (potasyum katyon-klorid cotransporter 3) ve CIP 1 (CCC interaktinprotein tip 1) mRNA fazla bulunurken KKC2 (Potasyum katyon klorid 2), CCC9 (katyonklorid cotransporter) düşük düzeylerde bulundu. KCC4 (Potasyum katyon kloridcotransporter 4) seviyesi değişmedi.Yargı: Bu sonuçlar bize bu modelde CCCsnin varlığının çok sıkı bir şekilde yazılımkontrolünde olduğu sonucunu gösterdi. Nöbete yol açan hipereksitabilite mekanizmasınıtetikleyen hücre içi ve hücre dışı klorid konsantrasyon dengesizliği bu deragülasyon sonucuolabilir. Bu durum beyinde epileptik nöbete yanıtta bir çeşit genetik programın aktiveolmasına bağlı olabilir ve hangi biyolojik yolağın module olacağını açıklayabilir.
Abstract
Objective: Intracellular chloride concentration of a given cell within the central nervoussystem is determined by a delicate balance between the Cl- extrusion system and the Cl-accumulation system. In this study, we aim to investigate the expression profiles of cation- chloride cotransporters (CCCs) mRNA in the mice model of lithium-pilocarpine inducedstatus epilepticus (PISE) and the possible relationships between CCCs and epileptogenesis.Methods: Male adult Balb/c mice were randomly divided into two groups: the PISE groupand the control. We used the mice of PISE group to establish the animal model of PISE andthe PISE group could be subdivided into three time points: 1 d, 14 d and 45 d. Real-timefluorescence quantitative PCR (polymerase chain reaction) was used to explore the expressionof CCCs family members mRNA in CA1 region.Results: These CCCs members all showed distinctive expression patterns. The expressionlevels of NKCC1 (sodium potassium cation-chloride cotransporter 1), KCC1 (potassiumcation-chloride cotransporter 1), KCC3 (potassium cation-chloride cotransporter 3) and CIP1(CCC-interacting protein type 1)mRNA were up-regulated while those of KCC2 (potassiumcation-chloride cotransporter 2) and CCC9 (cation-chloride cotransporter) mRNA were down- regulated after PISE. The expression of KCC4 (potassium cation-chloride cotransporter 4)remained unchanged.Conclusions: These data lead us to conclude that CCCs expression as a whole is under verystrict transcriptional control in this model. Deregulation of their expression might break thebalance of intracellular and extracellular chloride concentration which contributed to themechanism of hyperexcitability that led to seizures. This may be due to some form of geneticprogram activated in response to epileptic seizure in the brain and may highlight whichbiological pathways are modulated.
