Effects of Erythropoietin on The Volume of Infarction After Spinal Cord Ischemia Model

Özet

Giriş: Bu çalışmada, tavşan spinal kord iskemi modelinde 40 dakika iskeminin oluşturduğumotor nöron hücre infarktı ve infarkt alanına intravenöz eritropoietinin etkisini araştırmayıplanladık.Metod: Onyedi Yeni Zellanda tavşanı kullanıldı. Spinal kord iskemisi oluşturmak amacıylaabdominal aorta 40 dakika oklüde edildi. Tavşanlar randomize olarak 2 gruba ayrıldı. Kontrolgrubuna (n=7) serum fizyolojik, eritropoietin grubuna (n=7) 1000U/kg eritropoietin infüzyonuyapıldı. Motor fonksiyonlar 1., 24., 48. ve 72. saatte Drummond ve Moore kriterleri iledeğerlendirildi. Postoperatif üçüncü günde deneklere yüksek doz tiyopental verildi ve spinalkordları çıkarıldı. İskemik bölgelerdeki canlı motor nöron sayısı ve infarkt alanları kaydedildi.Motor fonksiyon ve infarkt alanları arasındaki farklar karşılaştırıldı.Bulgular: Eritropoietin, nörolojik fonksiyonları iyileştirmekle birlikte iskemik bölgelerdekimotor nöron infarktını ve infarkt alanını belirgin olarak azalttı (1,48 ± 0,69 % vs 2,00 ± 1,28% ).Sonuç: Eritropoietinin, 40 dakikalık spinal kord iskemisi sonrasında bile nörolojik hasarıazaltıcı etkileri klinik kullanımında spinal kord hasarı sonrası beklentileri iyileştirebilir.

Abstract

Background: In this study we aimed to investigate the effects of intravenous erythropoietinon motor neuron cells and volume of infarction in a rabbit model of spinal cord ischemia of40 minutes.Methods: Seventeen New Zealand Rabbits were used. Abdominal aorta was occluded for 40minutes to induce spinal cord ischemia. Subjects were randomly divided into 2 groups. In thecontrol group (n=7) we infused physiological saline solution while 1000 U/kg oferythropoietin in the group erythropoietin (n=7). Motor functions were evaluated with criteriaof Drummond and Moore at times 1., 24., 48., and 72th hours. At the third postoperative daysubjects were infused with high dose of thiopental and their spinal cords were removed. Thenumber of viable motor neurons and the volume of infarction were obtained in the ischemicregions. Comparisons regarding motor functions and infarct volumes revealed significantdifferences.Results: Erythropoietin treated animals not only neurological functions were improved, butalso number of motor neurons as well as the volume of infarction were decreased clearly inischemic regions (1,48 ± 0,69 % vs 2,00 ± 1,28 % ).Conclusion: Since erythropoietin seemed to protect the rabbits against neurologicaldeterioration even after 40 minutes of spinal cord ischemia, transferring these findings toclinical studies designed for spinal cord ischemia may improve the outcome expectations.
