Blocking The Activity of CTGF is Capable of Preventing The Development of Early- Stage Atherosclerosis in Rat Carotid Artery

Özet

Giriş: Atherosklerotik plakların oluşmasında bağ dokusu büyüme faktörü (BDBF)nün etkisibilinmekle birlikte ıraksak etkileri ancak tahmin edilmektedir. BDBFnin bloke edilmesi ileatherosklerozun oluşmasının önleneceği konusu ise kesinlik kazanmamıştır.Yöntemler: Yüksek oranda BDBF oluşturan erken-evre atheroskleroz oluşturan bir sıçankarotid arter atherosklerotik modeli oluşturduk. BDBF siRNA kullanarak vasküler düz kashücresinde BDBF oluşumunu durdurduk ve nötralize eden anti-BDBF antikoru enjekte ederekatherosklerotik lezyonda BDBF yapımını bloke ettik.Sonuçlar: BDBF aktivitesini bloke etmek arterial stenoz oranınında, intima ve tunica mediaalanınında artışı aşırı oranda önledi. Ayrıca atherosklerotik lezyon alanında MMP-2, MMP-9ve phospho-Akt proteinlerinde önemli ölçüde süpresyon oluşturdu. BDBF siRNA in vitroolarak vasküler düz kas hücresinde migrasyon ve büyümeyi süpresse etti.Yargı: BDBF erken-evre atherosklerozun gelişiminde önemli ölçüde etkilidir. BDBFninaktivitesinin bloke edilmesi Aktnin inaktivasyonuna ve MMP-2 ve MMP-9 un süpresyonunayol açan bir mekanizma ile erken-evre atherosklerozun gelişimini önleyici yeteneğe sahiptir.Bu sonuçlar atherosklerozun önlenmesinde yeni bir stratejiyi önermektedir.

Abstract

Background: Although connective tissue growth factor (CTGF) has been demonstrated tocontribute to the formation of atherosclerotic plaques, its divergent effects on atherosclerosishave been proposed. It is unclear if blocking the activity of CTGF may prevent thedevelopment of atherosclerosis.Methods: We established a rat carotid artery atherosclerotic model which recapitulates theearly-stage atherosclerotic lesion and highly expresses CTGF. We blocked CTGF expressionin atherosclerotic lesion by injecting a neutralizing anti-CTGF antibody and knocked-downCTGF expression in vascular smooth muscle cells (VSMCs) using a CTGF siRNA.Results: Blocking the activity of CTGF extensively prevented the increase in arterial stenosisrate, intima area, and tunica media, along with significant suppression of MMP-2, MMP-9,and phospho-Akt proteins in the areas of atherosclerotic lesions. CTGF siRNA suppressed thegrowth and migration of VSMCs in vitro.Conclusions: CTGF contributes significantly to the development of early-stageatherosclerosis. Blocking the activity of CTGF is capable of preventing the development ofearly-stage atherosclerosis through a mechanism involving the inactivation of Akt andsuppression of MMP-2 and MMP-9. Our result suggests a novel strategy to the prevention ofatherosclerosis.
